[0021] Example 1: Garlic: sterile single distilled water ratio of 2:1 (W / V), peeled garlic, mash into a slurry, add sterile single distilled water and stir for 2 minutes, then stand at room temperature for 30 minutes, filter and set aside.

Embodiment 2

[0022] Example 2: Garlic: sterile single distilled water ratio of 1:1 (W / V), peeled garlic, mash into a paste, add sterile single distilled water and stir for 2 minutes, then stand at room temperature for 30 minutes, filter and set aside.

Embodiment 3

[0023] Example 3: Garlic: sterile single distilled water ratio of 1:3 (W / V), peeled garlic, mash into a slurry, add sterile single distilled water and stir for 2 minutes, then stand at room temperature for 30 minutes, filter and set aside.

[0024] The above-mentioned garlic solution is used in the following ways:

[0025] At the end of winter, take the leaf buds of sweet cherry, peach, and Chinese rose as explants, bring them back indoors, and rinse them with tap water for 1 hour. Carry out explant sterilization under aseptic operation condition, put the leaf bud of sweet cherry, peach, Chinese rose into the garlic aqueous solution of embodiment 1, embodiment 2 respectively. Soak in medium for 30 minutes, take it out, rinse it with sterile single distilled water for 3 times, blot the water with sterile filter paper, and inoculate it into the primary medium (medium formula: MS+6-BA0.2mg / L+IBA0.05mg / L+ Agar 7g / L + sucrose 30g / L, pH5.8).

Abstract

The invention discloses an environment-friendly explant sterilizing solution, namely a garlic aqueous solution, which is prepared by peeling the garlic, mashing, and then mixing with sterile distilled water according to a weight ratio (by volume) of (1-9):1. The invention further provides an environment-friendly explant sterilizing method based on the sterilizing solution, wherein plant explant is immersed into the garlic aqueous solution for sterilizing. The new explant sterilizing method, namely the garlic aqueous solution method is moderate for treated plants, high in sterilization rate which can be up to 100%, stable in sterilizing effect, simple to operate, low in cost, and friendly to environment.

Description

technical field [0001] The present invention relates to the sterilization of plant explants, more specifically, to the improvement of the sterilization method and the sterilization solution. Background technique [0002] Explant sterilization is an important part of the success of plant tissue culture. Traditionally, during plant tissue culture, the commonly used explant sterilization method is: soak the explants in 70% or 75% ethanol for no more than 30s, and then use sodium hypochlorite (NaOCl) or calcium hypochlorite [Ca(OCl ) 2 ] or mercuric chloride (mercuric chloride, HgCl 2 ) to further sterilize, in order to receive a better explant sterilization effect. The usual concentration of mercuric chloride is 0.1%, and the treatment time is 10-15 minutes. The bactericidal effect is good, but the residual toxicity is high. After disinfection, it needs to be washed with sterile water for 3 to 5 times; the common concentration of sodium hypochlorite and calcium hypochlorite...
