The method is as below: first conducting smoothing filtering on collected immune chromatography fluorescence waveform to clear the fluorescence waveform; then conducting background threshold treatment on the waveform after filtering to remove the background signals in the waveform; and calculating a ratio of a detection line waveform envelope area and a quality control line waveform envelope area, conducting waveform width compensation and temperature compensation to finally obtain detection result of immune chromatography fluorescence. The invention uses a method of calculating envelope area ratio to realize automatic accurate detection of the immune chromatography fluorescence results; the twice compensation method combining the wave width compensation and temperature compensation to further improves the accuracy of immune chromatography fluorescence detection.
