The invention relates to a Dendrobium officinale propagation method which comprises the following steps: (1) adventitious bud induction: culturing a sterilized Dendrobium officinale stem section in an induction culture medium for 28-30 days, wherein the induction culture medium is composed of MS (Murashige-Skoog) basal culture medium, 0.1-0.15 mg/L naphthylacetic acid, 0.5-1.0 mg/L 6-benzylaminoadenine, 3 wt% sucrose and 0.8 wt% agar; (2) adventitious bud proliferation: after the induction is finished, cutting off the new bud growing at the internode of the stem section, inoculating into the induction culture medium to carry out subculture propagation for 30-32 days, and obtaining cluster buds at the cutting; and (3) sound seedling and root transplantation: cutting the cluster buds into single seedlings, transferring into a sound seedling culture medium to carry out sound seedling culture for 20 days, and carrying out rooting culture for 28 days after the buds develop into 2-3 blades which are 2-3cm high, wherein the sound seedling culture medium is composed of MS basal culture medium, 3 wt% sucrose, 0.7 wt% agar and 3 wt% organic fertilizer, and the rooting culture medium is composed of 1/2 MS basal culture medium, 0.3-0.5 mg/L naphthylacetic acid and 0.3-0.5 wt% activated carbon.
