Embodiment 1

[0044] Example 1: Isolation of filamentous fungus J-5 and test of its tolerance to heavy metals

[0045] 1. Strain isolation:

[0046] Weigh 10g of cadmium-contaminated soil into 90mL of sterilized normal saline, vibrate on a constant temperature shaker at 28°C at 150r / min for 15min, and after standing for 1min, take the supernatant for 10-fold serial dilution; then dilute 100μL of different dilution factors The solution was coated on a Martin Bengal red plate containing 10 mM cadmium, and placed upside down in a constant temperature incubator at 28°C for 5 days. The microorganisms grown on the plate were separated and purified by streaking, and then re-applied to the above-mentioned Martin Bengal Red plate containing cadmium for cultivation. After 10 times of isolation and screening, the strains with stable resistance to cadmium were preserved and stored on a PDA slant for future use.

[0047] The applicant named one of the strains isolated above as the filamentous fungus J...

Embodiment 2

[0054] Example 2: Experiment of cadmium adsorption capacity of filamentous fungus Penicillium chrysogenum J-5

[0055] 2.1 Static cadmium adsorption:

[0056] After the filamentous fungus J-5 was cultured on PDA medium for 5 days, the spores were collected and a spore suspension was prepared; then it was inoculated into 200 mL of fresh mineral salt medium at a ratio of 1:100 and cultured for 5 days, and the bacteria were collected by filtration and collected. Vacuum freeze-drying was carried out; then, the dried bacterial powder was passed through a sieve with a pore size of 300 μm for the analysis of cadmium adsorption.

[0057]Add 0.01 g of the above-mentioned pretreated bacterial powder into a 50 mL centrifuge tube containing 20 mL of cadmium solution, and then place the centrifuge tube in a constant temperature shaker at 28°C and maintain it at 150 r / min for 240 min. During the adsorption process, 100 μL of the supernatant was taken at 0, 15, 30, 45, 60, 120, 180 and 240 ...

Embodiment 3

[0060] Embodiment 3: Filamentous fungus J-5 and plant symbiotic hydroponic experiment

[0061] This example aims to demonstrate the ability of the filamentous fungus J-5 provided by the present invention to protect plant growth in simulated heavy metal polluted soil. See the literature (Green, 1989; Yu Fangming et al., 2008) for the planting and cultivation methods of pakchoi; the treatment method for the dry bacterial powder of the filamentous fungus J-5 is as described in Example 2 above. Before inoculating the filamentous fungus J-5 into the hydroponic system, collect the spores of the filamentous fungus J-5 and adjust the spore suspension to 10 7 pc / mL, spare.

[0062] Taking Chinese cabbage (Brassica chinensis L.) as the test plant, after accelerating germination, take 10 seeds with the same germination status and place them in a planting cup filled with 100g of sterilized vermiculite. After emergence, pour 10mL of sterilized tap water every 2 days, and change to Hoagla...

Abstract

The invention discloses high-cadmium-adsorption filamentous fungi penicillium chrysogenum J-5 as well as a preparation method and an application thereof. The preparation method comprises the following steps of: firstly weighing and putting 10g of cadmium polluted soil into 90mL of sterilized normal saline, vibrating on a constant-temperature shaking table, standing, taking supernatant liquid and diluting; secondly coating different dilution factors of diluent on a Martin rose-bengal flat plate containing 10mM of cadmium, and inversely placing the flat plate in a constant-temperature incubator for cultivation; thirdly, carrying out streaking and purification on microorganisms which grow on the flat plate, and then coating the organisms on the Martin rose-bengal flat plate containing cadmium again for cultivation; fourthly, after isolation and screening, storing a bacterial strain having stable resistance capacity on cadmium on a PDA (Potato Dextrose Agar) inclined plane. The obtained filamentous fungi penicillium chrysogenum J-5 has high tolerance and high adsorption property on multiple metals; the raw materials are easy to obtain and low in cost. The bacterial strain has relatively high fixation capacity on heavy metals in environments, can be used of reducing the biological toxic property of the heavy metals and is suitable for the in-situ remediation of large-area polluted soil.

Description

technical field [0001] The invention belongs to the field of environmental governance. More specifically, it relates to a preparation method of a filamentous fungus with high tolerance and high adsorption capacity to various heavy metals, and also relates to the use of a filamentous fungus Penicillium chrysogenum J-5. Background technique [0002] Cadmium (Cd) mainly exists in nature in the form of 13 minerals such as sulfur cadmium ore, and is often associated with other metal ores, such as lead-zinc ore. The average concentration of Cd in the earth's crust is 0.15mg / Kg (Yan Mingjuan, 1999), the Cd concentration in soil is 0.01-2mg / Kg, and the Cd concentration in various water bodies is 0.11-10μg / Kg. Although under normal circumstances, Cd exists in a trace amount in the natural environment, but human activities have increased its content and distribution in the environment. For example, Cd can directly enter the atmosphere, water body and soil, and migrate in different e...
