[0031] SD大鼠40只,体重220-280g,购自第四军医大学动物实验中心。动物分笼饲养(环境温度24℃,湿度60-70%),术前6d禁食、自由饮水。10%水合氯醛腹腔注射麻醉,仰卧固定,链接心电图检测,气管插管并接呼吸机(频次:75次 / min ;潮气量:8-9 mL;吸:呼=1:2),乳香乳剂静脉注射(3各剂量水平200,100,50 mg / kg),30 min后眼眶采血1.0mL。给药30 min后,胸骨左缘旁纵行切开,第3-5肋间做横向切口,挑出心脏,以左心耳与肺动脉圆锥交界处的左冠状动脉前降支(Left anterior descending coronary aery, LAD)为标志,于左心耳下方1-2mm处进针,穿线(横跨左冠状动脉前降支),结扎冠状动脉前降支血管30 min后再灌注4h时取血测定CK-MB和LDH活性,取心脏组织测定心肌梗死面积。结果(见表1):高中低三个剂量乳香乳剂均能显著预防缺血 / 再灌注造成的心肌损伤,与模型组相比可明显提高CK-MB和LDH活性,同时减小心肌梗死面积( figure 1 ),表明乳香乳剂具有心肌损伤保护作用。
