Embodiment 1

[0024] 1. Preparation of reference solution

[0025] Take 10mg of rutin reference substance, weigh it accurately, put it in a 50ml volumetric flask, dissolve it with an appropriate amount of 70% ethanol, dilute to the mark, shake well, and get it (every 1ml contains 0.2mg of rutin).

[0026] 2. Preparation of the test solution

[0027] Accurately weigh 3.0g of Qumai powder, put it into a round bottom flask, add 30ml of petroleum ether, reflux for 3.0h in a water bath at 70°C, filter the dregs, evaporate the solvent, add it to a 100ml Erlenmeyer flask, add ethanol solution, and ultrasonically extract. Suction filtration after the completion of ultrasound, discard the dregs, transfer the filtrate to a 100ml volumetric flask, dilute to volume with 70% ethanol, and set aside. According to the preliminary test, ethanol volume fraction, ultrasonic time, ultrasonic power, and solid-liquid ratio were selected as the investigation factors, and three levels were determined on the basis...

Embodiment 2

[0035] 1. Single factor experiment

[0036] According to the preparation method of the test solution, the single factor experiment of different ethanol concentration, solid-liquid ratio, ultrasonic time and ultrasonic power was carried out, and the relationship between the extraction effect of total flavonoids and the factor level was obtained.

[0037] 1.1 The effect of ethanol concentration on the extraction rate of total flavonoids of Qumai

[0038] Accurately weigh 3.0g of Qumai powder, the ratio of solid to liquid is 1:20, the ultrasonic power is 150W, and the ultrasonic is 40min, and the concentration is 30%, 40%, 50%, 60%, 70%, 80% ethanol for ultrasonic extraction respectively, according to The assay method described in Example 1 measures the absorbance, the results are shown in Table 2, and the total flavonoid content is calculated.

[0039] 1.2 The effect of extraction time on the extraction rate of total flavonoids of Qumai

[0040]Accurately weigh 3.0g of Qumai p...

Embodiment 3

[0069] Carry out a series of technological verification experiments on the above-mentioned quince flavonoid extraction process:

[0070] 1. Inspection of precision

[0071] Precisely measure 6 parts of the same test solution, each 2.0ml, place in a 25ml volumetric flask, develop color according to the aforementioned color development method, take the corresponding reagent as a blank, measure the absorbance at 510nm, and calculate the total flavonoids by regression equation content, the results are shown in Table 9. The RSD was 0.29%, indicating good precision.

[0073]

[0074] 2. Stability inspection

[0075] Accurately measure the same test solution, develop color according to the aforementioned color development method, measure the absorbance once every 5 minutes at 510nm, measure for 60 minutes, the results show that the absorbance of the test solution is stable within 30 minutes, the results are shown in Table 10 Shown, RSD is 3...

Abstract

The invention provides an ultrasonic extraction method for extracting flavone from dianthus superbus. According to the ultrasonic extraction method, the dianthus superbus is taken as a raw material and ultrasonic wave is adopted for treating crushed dianthus superbus cells to obtain dianthus superbus flavone with high purity. In an experiment, total flavone content is selected as an evaluation index to obtain the optimal ultrasonic extraction process of Mongolian dianthus superbus; an ultraviolet-visible spectrophotometry is adopted for determining the total flavone in the dianthus superbus and the content of the total flavone in the dianthus superbus is 0.6367%; and the ultrasonic extraction method has the advantages of simple operation method, and good stability, precision, repeatability and sample feeding and recycling rates. The ultrasonic extraction method is good for improving the extraction rate of the flavone, shortening the extraction time and saving an extraction solvent; and the method can be used for evaluating the quality of the Mongolian dianthus superbus.

Description

technical field [0001] The invention relates to the technical field of bioengineering, in particular to an ultrasonic extraction method for extracting flavonoids from fenugreek. Background technique [0002] Qumai, also known as wild wheat, rock osmanthus, giant sentence wheat, etc., is the flowered aerial part of Dianthus superbus L., which is distributed in North Korea, Mongolia, Russia, Northeast China, North China, Northwest China, East China, Sichuan, Guizhou and other places. The aerial part of Qumai with flowers is used as Mongolian medicine. Qumai is bitter in taste, cold in nature, blunt, light and thin, cools blood, relieves pain and detoxifies. Qumai treats blood heat, tingling pain due to blood heat, liver heat, "Bao Ri" heat and cold mixed period, Sha syndrome, postpartum fever, and has the effects of diuresis and menstrual flow, breaking blood and menstruation. It has a wide range of clinical curative effects and can be used as a compound or alone. It is a co...
