[0024] method

[0025] Human stool sample collection. Spanish subjects were healthy controls or patients with chronic inflammatory bowel disease (Crohn's disease or ulcerative colitis) in clinical remission. Patients and healthy controls were asked to provide frozen stool samples. Obtain fresh stool samples at home and immediately freeze the samples by storing them in their home freezer. Frozen samples were delivered to the hospital using closed polystyrene foam containers, and they were then stored at -80°C until analysis.

[0026] DNA extraction. Frozen aliquots (200 mg) of each stool sample were suspended in 250 μl of guanidine isothiocyanate, 0.1 M Tris (pH 7.5) and 40 μl of 10% N-lauroyl sarcosine. DNA extraction was then performed as previously described (Manichanh et al., Gut, 55:205-211, 2006). The concentration of DNA and its molecular size were estimated by NanoDrop (Thermo Scientific) and agarose gel electrophoresis.

Abstract

A new method for diagnosing an inflammatory bowel disease is herein described, based on the determination of the absence of at least one gene from the human' gut microbiome.

Description

technical field [0001] Inflammatory bowel disease is a chronic disease of unknown etiology characterized by persistent mucosal inflammation at varying levels in the gastrointestinal tract. Ulcerative colitis and Crohn's disease are the two main types of inflammatory bowel disease. Ulcerative colitis causes persistent mucosal inflammation limited to the colon, whereas Crohn's disease causes non-persistent transmural inflammation anywhere throughout the GI tract, although it most commonly affects the terminal ileum ( terminal ileum). The most common intestinal lesions consist of mucosal ulceration, swelling of the intestinal wall, and stricturing of the intestinal lumen. These chronic inflammatory lesions can lead to symptoms such as diarrhea, faecal urgency, abdominal pain and fever, and complications of varying severity including bleeding, intestinal obstruction, sepsis and malnutrition. Background technique [0002] Epidemiological studies have demonstrated a steady incr...
