Embodiment Construction

[0040] Some preferred embodiments of the invention are described in detail here by way of non-limiting examples.

[0041] In the following description numerous specific details are given to provide a thorough understanding of the embodiments. Embodiments may be made with or without these specific details, or with other methods, components, materials, and the like. In other instances, well-known structures, materials, or operations are not shown or described in detail to avoid obscuring the embodiments.

[0042] The headings given here are for convenience only and are not intended to interpret the scope or meaning of the embodiments.

[0043] In addition to damaging intracellular targets, ionizing radiation (mostly by generating reactive oxygen species) also modulates the activity of regulatory molecules that control stress and restorative biological responses.

[0044] In this response, transcriptional upregulation of the MET oncogene appears to be a critical event leading t...

Abstract

The invention relates to MET inhibitors for enhancing radiotherapy efficacy. Met inhibitor and / or nucleotide sequence encoding an MET inhibitor for use in the treatment of patients suffering from a cancer for reducing and / or abrogating patients' resistance to radiotherapy, wherein the Met inhibitor is selected among: i) an anti-Met monoclonal antibody, ii) a genetically engineered antibody containing the complementarity determining regions (CDRs) of the anti-Met monoclonal antibody, and iii) a fragment of (i) or (ii) containing the complementarity determining regions (CDRs) of the anti-Met monoclonal antibody, or combinations thereof.

Description

field of invention [0001] This specification relates to the use of MET inhibitors in enhancing the efficacy of radiotherapy for tumor patients. technical background [0002] Although radiotherapy has been successfully used to treat cancer patients, it may fail to eradicate tumors, which recur with a more aggressive phenotype. Consistent with this, classical studies in animal models have revealed paradoxical pro-metastatic effects of ionizing radiation (IR). Tumor progression after radiation therapy may be due to positive selection of a subpopulation of "cancer stem cells", which are inherently radiation resistant. However, significant evidence suggests that, in addition to selection, IR promotes an adaptive phenotype for tissue regeneration that may manifest as metastatic behavior. This phenotype is defined as a "stress-and-recovery" response to DNA damage that occurs at both the single-cell and tissue levels. In single cells, the detection of DNA damage yielded specific ...
