[0015] Field Trial Design and Sampling

[0016] The field experiment was carried out in the seedling base of Jiande City, Zhejiang Province. The experimental field was divided into 9 plots, and the three rice varieties were planted respectively. Each variety had 3 repeated plots, and each plot had 26-28 rows×12 plants / row≥ There are 300 plants, and the plots of different varieties are set at intervals.

[0017] Sampling was performed at the tillering, heading, filling and maturity stages of rice. The rice and the soil at its roots were pulled out with a spatula and brought back to the laboratory, and three rice samples were taken from each plot. After the retrieved samples were moderately air-dried at room temperature, the soil around the rhizosphere was shaken off and the soil around the rhizosphere was collected. After removing debris such as plant roots and gravel, the soi...

Abstract

The invention discloses a method for analyzing breathing intensity of soil microorganisms in Bt rice plantation and straw returning. The method comprises the following steps of: mixing an acquired soil sample and a glucose solution uniformly in a sealed container, culturing for 7 and 24 hours respectively, and measuring the amount of CO2 generated by each gram of soil sample by using gas chromatography, wherein the amount of the CO2 respectively represents the breathing intensity of the soil microorganisms in Bt rice plantation and straw returning and the capacity of soil microorganisms of utilizing glucose. The invention has the advantages that: the method is simple and quick; and most importantly, the amount of CO2 generated in 7 hours and 24 hours can be used for identifying the influence and the restoring force of Bt rice plantation and straw returning on the soil microorganisms.

Description

technical field [0001] The invention belongs to the field of ecological environment protection, and in particular relates to a method for analyzing the respiration intensity of soil microorganisms suitable for Bt rice planting and straw returning to the field. Background technique [0002] In order to overcome the disadvantages caused by the extensive use of pesticides and ensure high and stable yields of crops, the research and development of genetically modified crops has developed rapidly. In 2009, the global area of genetically modified crops increased from 0.174 billion hectares in 1996 to 134 million hectares. Transgenic crops of insecticidal toxic gene fragments of Bacillus thueingiensis have resistance to Lepidoptera, Diptera, Hymenoptera, Homoptera, Orthoptera ), Mallophage and other insects and their larvae, and it is also one of the important transgenic crops in my country. However, whether the use of genetically modified crops poses a potential threat to farmlan...
