Problems solved by technology

[0007] However, no other BMP proteins have been reported to be involved in melanocyte biology such as melanogenesis and / or melanin transfer
Although much is known about the regulation of melanogenesis (i.e., melanin synthesis) in melanocytes, our knowledge of how melanosomes are transferred from melanocytes to keratinocytes is limited

[0110] introduction

[0111] In a study of the biology of melanin transfer, the present inventors investigated the potential role of members of the bone morphogenetic protein (BMP) family in this process. In this application a novel mechanism controlling the transfer of melanin from human skin melanocytes to human skin keratinocytes is described. This includes the action of BMP6, which can stimulate this process in a dose-dependent manner. Moreover, a selective BMP6 antagonist (sclerostin) inhibits BMP6-induced melanosome transfer and blocks the production of BMP6-induced nanotubes (called filopodia), as scanned for human melanocytes Evidenced by electron microscopy (SEM) analysis. Carefully dissecting the molecular mechanism of BMP6-induced filopodia formation, using reverse transcription analysis, it was found that BMP6 stimulated (cell proliferation cycle 42) the expression of Cdc42 in human melanocytes in a dose-dependent manner. SEM analysis of FM55 cells transfected w...

Abstract

The present invention relates to compositions and methods useful in studying or modulating melanin pigmentation in the skin. Particularly, the invention relates to compositions comprising a substance capable of modulating the activity or expression of ALK6 (SEQ ID 2) or Cdc42 which in turn are capable of modulation of the transfer of melanin from melanocytes to keratinocytes and potentially from keratinocytes to keratinocytes. The invention also relates to assays for identifying such compositions, and methods of modulating skin pigmentation.

Description

technical field [0001] The present invention relates to compositions and methods useful for studying or modulating melanin pigmentation in the skin. Specifically but not exclusively, the present invention relates to compositions capable of modulating the transfer of melanin from melanocytes to keratinocytes and potentially from keratinocytes to keratinocytes, assays for identifying such compositions, and modulating Approaches to skin pigmentation. Background technique [0002] Melanocytes are neural crest-derived cells distributed along the basal layer of the epidermis and hair bulb. These cells synthesize melanin inside melanocyte-specific organelles called melanosomes. For hyperpigmented skin and hair, this melanin must be transferred from melanocytes to adjacent keratinocytes. The quantity and quality of melanin (brown / melanin eumelanin or red / xanthomelanin) determine mammalian skin and hair / wool / The color of the fur. In the past two decades, much progress has been ...
