Problems solved by technology

However, most of the research on poly-γ-glutamic acid produced by microbial fermentation is still in the laboratory stage, and its purity needs to be further improved when used as a biological material [Bajaj I B, Lele S S, Singhal R S, Biores Tech, 2009, 100:826-832]

Embodiment 1

[0023] (1) Weigh 0.40 g of sodium alginate and dissolve it in 10 mL of phosphate buffer solution (pH=5), and stir at room temperature until the sodium alginate is completely dissolved.

[0024] (2) Add 0.87 g of EDC to the solution (1), and stir at 4° C. for 30 min.

[0025] (3) Add 0.28 g of cystamine to the solution (2) as a cross-linking agent, stir and fully dissolve, pour the reaction solution into a plastic culture plate, and place it at room temperature for 3 hours to form a hydrogel.

[0026] (4) The hydrogel obtained in step (3) was pre-frozen in a refrigerator at -50°C for 24 hours, and then freeze-dried for 2 days to obtain a porous scaffold material. The swelling rate of this unmodified three-dimensional porous scaffold in deionized water is about 35g / g, and the degradation time in 50mM cysteine solution is about 15h.

[0027] (5) Soak the porous scaffold obtained in step (4) in a 3 mg / mL chitosan (molecular weight 5k) solution for 4 hours, so that the chitosan ...

Embodiment 2

[0030] Raw material, preparation method are substantially identical with embodiment 1. It is 0.56g that the step (3) among the embodiment 1 is adjusted to cystamine addition, and the chitosan molecular weight in the step (5) is adjusted to 10k, and other steps are identical. The swelling ratio of the unmodified three-dimensional porous scaffold is about 24g / g. The swelling rate of the three-dimensional porous scaffold modified by chitosan solution was 6g / g.

Embodiment 3

[0032] Raw material, preparation method are substantially identical with embodiment 1. Step (1) in Example 1 was adjusted to an addition of sodium alginate of 0.45 g, and step (2) was adjusted to an addition of EDC of 0.96 g. The unmodified 3D porous scaffold has low strength and can barely maintain its shape with a swelling rate of about 29 g / g. However, the compressive strength of the modified three-dimensional porous scaffold can reach 4.5±1.3kPa, the compressive modulus is about 27.4±6.1kPa, and the swelling rate is 8g / g.

Abstract

The invention relates to a chitosan modified alginate hydrogel three-dimensional porous bracket with specific in-vitro degradability and a preparation method thereof. The method comprises the following steps: dissolving sodium alga acid serving as a raw material in phosphate buffer solution; performing amidation reaction of a carboxyl group in the sodium alga acid and an amino group in a cross-linking agent cystamine or dimethyl cystinate under the activation of water-soluble carbodiimide to form a chemically crosslinked hydrogel; performing freeze drying on the hydrogel to obtain a porous bracket material of the hydrogel; and performing surface modification on the porous bracket by using chitosan. In solution of a reducing agent such as cysteine with an appropriate concentration, a disulfide bond in a hydrogel cross-bridge is degraded through a disulfide bond-sulfydryl conversion reaction, so the porous bracket is decomposed and dissolved and disappears. Therefore, the porous bracketcan be used as an in-vitro cell culture template material. The hydrogel porous bracket researched by the invention has the characteristics of simple preparation, rich raw material source, low cost and availability. Various physiochemical performances, mechanical strength, degradation rate and surface properties of the bracket material are controllable within a large range.

Description

technical field [0001] The invention relates to the manufacture of a three-dimensional porous template material for tissue construction, in particular to a chitosan-modified alginate hydrogel three-dimensional porous scaffold and a preparation method thereof, specifically a specific in vitro degradable three-dimensional alginate hydrogel Porous scaffolds and methods for their preparation. Background technique [0002] Tissue engineering uses the principles and methods of life science and engineering science to research and develop a new generation of clinically applied substitutes that can repair or improve the functions of human tissues or organs, and are used to replace part or all of the functions of tissues or organs. The development of tissue engineering poses new challenges to biomaterials. In the past, the research and development of biomaterials were mostly conceived from the chemical, physical and processing properties of materials, and did not design from the cellu...
