Embodiment Construction

[0043] Normal adult Sprague-Dawley rats were selected, and proton magnetic resonance spectroscopy and microdialysis high-performance liquid chromatography were performed after one week of observation.

[0044] 1. Proton Magnetic Resonance Spectroscopy Analysis

[0045] Rats were continuously anesthetized with 1.5% isoflurane (laughing gas to oxygen ratio: 70:30) through the nose and placed in a special rat experimental bracket. The anal temperature was maintained at 37.0-37.5°C, and the head was fixed. Z The positive direction of the axis corresponds to the front end of the rat brain, and it is scanned with an animal-specific nuclear magnetic resonance imager-German Bruker Biospec 47 / 30, whose superconducting magnet magnetic field strength is 4.7T, by selecting the rat region of interest (Region of interest, ROI), the parameters of live body localization: repetition time (TR) = 2.5s, echo time (TE) = 136ms, average number of times (NA) = 256, after 1 H MRS acquisition of rat ...

Abstract

The invention provides a detection method for obtaining rat sinkaline signals through Proton MR spectroscopy (<1>H MRS) and then indirectly reflecting the content of acetylcholines in a rat. The method compares the content relationship of total sinkaline and acetylcholines in a sea horse area, a striaturn area, a frontal cortex area and a somatic cortex area of the normal rat; the contents of the total sinkaline and the acetylcholines of the rat in the four brain areas have obvious relativity through double variant and linear regression analysis. As shown by the result, the rat sinkaline signals obtained through the Proton MR spectroscopy are used as the predicated value of the content of the acetylcholines in the rat, which has statistic significance. Simultaneously, loss of the acetylcholines is a very important neuropathological variation of presenile dementia, thereby the method is hopeful to be applied to noninvasive early clinical diagnosis of the presenile dementia.

Description

technical field [0001] The invention belongs to the technical field of medical evaluation and detection. It specifically relates to the establishment of an early and non-invasive diagnosis method for Alzheimer's disease. Background technique [0002] Alzheimer's disease (AD) is a progressive neurodegenerative disease. With the acceleration of the aging process of society, the incidence and number of AD have risen sharply, and it has become the fourth leading cause of death after cardiovascular disease, cancer and stroke. However, so far, the etiology and pathogenesis of AD have not been fully elucidated. The current diagnosis of AD mainly relies on psychological scales, and its diagnosis depends on neuropathological examination. Because biopsy is traumatic to patients, it is not easy to be accepted by patients and their families. Therefore, the vast majority of patients can only be diagnosed by autopsy. On the other hand, new treatment methods are constantly emerging, and...
