AI Technical Summary

Benefits of technology

The present invention relates to a modified cell with targeted effector activity that can be used for the treatment of immune responses and tumors or cancers. The modified cell has a chimeric antigen receptor (CAR) that targets specific antigens on target cells. The CAR can be introduced into monocytes, macrophages, or dendritic cells, which then express the CAR and possess targeted effector activity. The modified cell can also be genetically modified to express the CAR. The invention also includes a pharmaceutical composition comprising the modified cell and a method of treating diseases or conditions associated with tumors or cancers by administering the modified cell to a subject. The technical effects of the invention include improved targeted effector activity and enhanced immune response against tumors or cancers.

Problems solved by technology

Despite the high response rates demonstrated in hematopoietic malignancies, CAR T cell efficacy in solid tumors (as well as in certain lymphoid tumors) may be limited.

Abstract

The present invention includes methods and compositions for treating cancer, whether a solid tumor or a hematologic malignancy. By expressing a chimeric antigen receptor in a monocyte, macrophage or dendritic cell, the modified cell is recruited to the tumor microenvironment where it acts as a potent immune effector by infiltrating the tumor and killing the target cells. One aspect includes a modified cell and pharmaceutical compositions comprising the modified cell for adoptive cell therapy and treating a disease or condition associated with immunosuppression.

Description

CROSS-REFERENCE TO RELATED APPLICATION[0001]The present application is a 35 U.S.C. § 371 national phase application from, and claims priority to, International Application No. PCT / US2016 / 044440, filed Jul. 28, 2016, and published under PCT Article 21(2) in English, which is entitled to priority under 35 U.S.C. § 119(e) to U.S. Provisional Application No. 62 / 197,675, filed Jul. 28, 2015, all of which are incorporated herein by reference in their entireties.BACKGROUND OF THE INVENTION[0002]Cancer immunotherapy has demonstrated exciting clinical results in the setting of numerous solid tumors and hematologic malignancies. The endogenous immune system is typically non-reactive to malignant cells, or can be actively immunosuppressive with respect to the body's reaction to the presence of malignant cells. One way to enhance treatment of tumors is to force tumor recognition by the immune system through genetic engineering of leukocytes. T cells can be engineered to express a synthetic immu...
