example 1

[0022] Example 1: please refer to the preparation process of the present invention figure 1 . The yeast GS115 in the logarithmic growth phase of the suspension culture was centrifuged to collect the bacteria, and mixed evenly with 15g / L sodium alginate solution. Under the action of a high-voltage electric field of a high-power microcapsule preparation instrument, drop into 100mmol / L calcium chloride solution for gelation reaction for 30 minutes to prepare calcium alginate gel beads with a particle size of 300μm, and then mix with 0.5g / L Polylysine solution film-forming reaction for 10 minutes, 55mmol / L sodium citrate solution was liquefied for 10 minutes after washing with normal saline for 3 times, and polylysine microencapsulated yeast was prepared after washing with normal saline for 3 times. 25 μm. The whole preparation process is completed under physiological conditions, and the survival rate of yeast cells is maintained at 100% during the preparation process.

example 2

[0023] Example 2: Put the prepared sodium alginate-polylysine microcapsules and calcium alginate microgel beads containing yeast cells in the YPD culture medium, culture in a shaker flask, 28°C, 180rpm, after culturing for 24 hours, calculate Yeast Cell Leakage Rate. The results showed that the cell leakage rate of the polylysine microcapsule group was more than 3 times lower than that of the calcium alginate microgel bead group.

example 3

[0024] Example 3: figure 2 The diffusion curves of proteins with different molecular weights in sodium alginate-polylysine microcapsules are given. It can be judged from the figure that molecules below 10,000 Da can freely diffuse in sodium alginate-polylysine microcapsules. It does not affect the supply of nutrients and the discharge of metabolites of the yeast in the capsule.

Abstract

The present invention relates to immobilized yeast cell sodium alginate-polylysine microcapsule preparing process. On the basis of original immobilized alginate cell, one great power microcapsule preparing instrument is adopted to make the sodium alginate solution with homogeneously mixed yeast cell under high voltage electrostatic field form homogeneously distributed liquid drops of controllable particle size in 100-500 micron and form micro bead with fixing calcium solution. The surface of the micro bead has one layer of microcapsule polylysine film formed via ion complexing reaction, and calcium in calcium alginate gel is replaced with sodium citrate solution to form liquefied liquid environment inside the microcapsule. The present invention is prepared under mild completely physiological condition of pH 5.0-7.4, 4-30 deg.c and no shearing and vibration. The preparation process has yeast cell activity 100 % maintained and the culture process has cell leakage less than 10 %.

Description

technical field [0001] The invention relates to a method for preparing microcapsules, in particular to a method for preparing microcapsules with immobilized yeast cells. Background technique [0002] Cell immobilization technology is an emerging biotechnology developed as early as the 1970s [Document 1: Kierstan M, Bucke C. The immobilization of microbial cells, subcellular organelles, and enzymes in calcium alginate gels. BiotechnolBioeng 1977 Mar; 19(3):387-97], which is one of the important contents in the field of modern bioengineering. Its essence is to use physical or chemical methods to fix free cells in a certain space and use them continuously as biocatalysts. The advantages of cell immobilization technology over free cells are mainly reflected in: (1) the concentration of cells in the reactor can be increased, the reaction speed is accelerated, and the volumetric production efficiency is improved; (2) cells can be used continuously; (3) strong anti-pollution abili...
