Embodiment 1

[0085] The tested variety: Xinhai 18, the main variety of sea-island cotton popularized in Xinjiang, can be obtained from the seed management station of Xinjiang Uygur Autonomous Region.

[0086] Medium: Prepare seedling medium, induction medium, elongation medium and rooting medium according to conventional methods. Their specific components and contents are:

[0087] Seedling medium: 1 / 2MS medium, in which agar 6g / L, glucose 30g / L, the pH value of the medium was adjusted to 5.8.

[0088] Induction medium: containing the macroelements, trace elements and organic components of MS medium at the concentrations shown in Table 1, and adding 2.5mg / L6-BA, glucose 30g / L, and curing agent Gelrite 2.4g / L. The pH was adjusted to 5.8.

[0089] Elongation medium: containing the macroelements, trace elements and organic components of MS medium at the concentrations shown in Table 1, and adding 6-BA2.0mg / L and GA 3 1.5mg / L, glucose 30g / L, and curing agent Gelrite 2.4g / L, the pH value of ...

Embodiment 2

[0093] The tested variety: Xinhai No. 16, the main variety of sea-island cotton popularized in Xinjiang, can be obtained from the seed management station of Xinjiang Uygur Autonomous Region.

[0094] As in Example 1, the required various media were prepared according to conventional methods, except that the concentration of the hormone 6-BA in the induction medium was 2.0 mg / L; the hormone and content in the elongation medium were 6-BA 0.4 mg / L and IAA0.5mg / L. According to the method described in Example 1, cotton seeds of Xinhai No. 16 were subjected to tissue culture experiments, and the experimental results were counted to calculate the regeneration frequency (results are shown in Table 2).

Embodiment 3

[0096] The variety tested: Xinhai 15, the main variety of sea-island cotton popularized in Xinjiang, can be obtained from the seed management station of Xinjiang Uygur Autonomous Region.

[0097] As in Example 1, prepare the required various media according to conventional methods, except that the hormone and concentration of the induction medium are 6-BA4.0mg / L; the hormone and content of the elongation medium are 6-BA0.4mg / L +IAA0.5mg / L. According to the method described in Example 1, tissue culture experiments were carried out on Xinhai No. 15 cotton seeds, and the experimental results were counted to calculate the regeneration frequency (results are shown in Table 2).

[0098] In the above-mentioned embodiment of the three sea-island cotton varieties, the regeneration frequency is shown in Table 2 below.

Abstract

The invention relates to a method that utilizing the cotyledon stalk of Gossypium barbadense( G. barbadense L. ) as explant to produce regenerative plants by tissue culture. The seeds of Gossypium barbadense disinfected are laid on the culture medium of seed to germinate, and the cut cotyledon stalks are laid in the inducing culture medium, and grown clump buds are cut to lay in the stretching culture medium for stretching, afterwards transferred into culture medium for growing root, to get regenerative plants. The inventive method can produce tufted buds that clumping and have no limitation of genetic types between every kind, short period of cultivation, normal growth of regenerative plants. The regenerative system of tufted buds of Gossypium barbadense cotyledon stalks established by said inventive method is suitable for every kind of Gossypium barbadense.

Description

technical field [0001] The invention relates to a method for tissue culture of sea-island cotton (Gossypium barbadense L.) using cotyledon petioles as explants for producing cotton regenerated plants. The cotton tissue culture method can also be applied to the genetic transformation of cotton. Background technique [0002] Cotton (Gossypium L.) is an important economic crop in the world and the most widely used natural fiber in the textile industry. Its annual output worldwide exceeds 100 million bales, worth 45 billion US dollars. The use of biotechnology to create new cotton germplasm, and then to breed new cotton varieties with improved traits is a supporting technology for the sustainable development of modern agriculture. Therefore, the research on the application of modern biotechnology to cotton has been widely valued, and the research on cotton tissue culture is an extremely important aspect. Tissue culture is the basis of biotechnology, that is, whether biotechnol...
