Embodiment 1

[0035] see figure 1 , the present invention provides a method for developing a group of mung bean InDel molecular markers, the method is used to provide a mung bean InDel molecular marker that the existing mung bean molecular markers do not have, and the molecular markers are aimed at insertion / deletion between genomes of different parents Marks designed for differences. In this embodiment, the method for developing the mung bean InDel molecular marker and its application includes the following steps, that is, steps (1)-(6).

[0036] (1) Select mung bean parents to cross, and use the hybrid offspring to construct mung bean genetic mapping population, that is, select appropriate parents to cross and construct mapping population. In this example, the male parent is Weilu No. 11, which is a mung bean variety with dwarf and medium-grained purple stems. The female parent is Sulu 16-10, which is a mung bean variety with tall stalks and large green stems. The parents are harvested ...

Embodiment 2

[0064] This embodiment provides a set of mung bean InDel molecular markers, which are obtained through the method for developing mung bean InDel molecular markers in Example 1. In this embodiment, the mung bean InDel molecular marker is obtained by PCR amplification using any one or several primer pairs shown in Table 1 in Example 1.

[0065] The mung bean InDel molecular marker is an InDel marker, which has the advantages of abundant quantity, stable genetics, good repeatability, and simple operation, and is an InDel marker with good polymorphism, which can be used for the analysis of mung bean genetic diversity, the construction of genetic linkage maps and Target gene mapping and molecular marker-assisted breeding provide InDel markers to make up for the lack of InDel markers in mung bean.

Embodiment 3

[0067] This example provides a mung bean hybridization method, which can be applied to obtain the mung bean genetic mapping population in Example 1. Wherein, the mung bean hybridization method comprises the following steps.

[0068] 1. Two mung bean parents are provided, one parent is used as the male parent, and the other parent is used as the female parent. These two mung bean parents can adopt Weilu No. 11 and Sulu 16-10 used in Example 1, and other mung bean varieties can also be used. The specific species can be selected according to actual needs, but the comparison of the differences in agronomic traits between the mung bean parents is guaranteed. Large, in order to facilitate subsequent identification of phenotypes and QTL loci.

[0069] 2. According to the flowering period of the mung bean parents, adjust the sowing time of the male and female parents or sow in stages so that the flowering periods of the male and female parents meet. Here, since the flowering period ...

Abstract

The invention discloses a set of mung bean InDel molecular markers and a development method thereof. The development method comprises the steps: mung bean parents are selected to be hybridized so as to construct a mung bean genetic mapping population; parent genomes are re-sequenced; parental sequencing data are subjected to quality control and then compared to mung bean reference genomes to screen differential sequences, then InDel loci between the parental genomes are screened, finally primers are designed for the InDel loci, and the multiple pairs of primers are obtained; the polymorphic primers evenly covering 11 chromosomes of mung beans are selected, and a plurality of filial generation plants are selected at random to construct a mung bean genetic linkage map; and the hundred-grainweight per plant is measured, quantitative trait loci (QTL) for controlling and adjusting the hundred-grain weight are identified, and QTL intervals and the linked markers are obtained. The success rate and the polymorphic primer ratio of primer design are increased, polymorphism is better, and the genetic linkage map constructed through the InDel markers can be used for identification of the QTLof quantitative traits and is beneficial to important gene identification of the mung beans and assistant breeding of the molecular markers.

Description

technical field [0001] The invention relates to a method for developing molecular markers in the technical fields of molecular biology, genetics and genomics, in particular to a method for developing a group of mung bean InDel molecular markers, and also relates to a group of mung bean InDel molecular markers developed by the method. Background technique [0002] Mung bean is a cultivated species in the genus Vigna of the leguminous bean family. It is rich in nutrients and has a grain protein content as high as 19.5% to 33.1%, which is higher than that of rice, wheat, corn and other cereal crops. It is a high-protein, medium-starch, and low-fat food. . Not only that, mung beans are also rich in a variety of mineral elements, vitamins and active substances. They have the functions of detoxification, antibacterial and antibacterial, anti-allergic, lowering blood fat, lowering blood pressure, anti-tumor, and preventing cancer. They are medical and food dual-purpose crops. The ...
