Embodiment 1

[0034] Example 1: Effect of FG-4592 on Migration of Epidermal Stem Cells

[0035] Experimental design: GF-4592 was purchased from Selleckchem, with a purity of 99.27%. The experiment was divided into a control group and a GF-4592 group. The epidermal stem cells of 1-day-old mice were isolated and cultured by classical two-step digestion method and differential attachment method, and the purity of epidermal stem cells was identified as 95% by CD71 and CD49f flow cytometry.

[0036]The epidermal stem cells were inoculated in a 24-well plate, and when the primary epidermal stem cells reached 70% confluency, administration was performed. The control group was added with 0.1% DMSO (dimethyl sulfoxide), and the GF-4592 group was added with a working concentration of 10 μM FG- 4592, after 24 hours of treatment, put them into the living cell workstation, randomly select 3-5 fields of view for each group, observe continuously for 3 hours, and set it to take pictures every 5 minutes to ...

Embodiment 2

[0039] Example 2: Effect of FG-4592 on the proliferation of epidermal stem cells (1)

[0040] Experimental Design: The experiment was divided into control group and GF-4592 group. The epidermal stem cells of 1-day-old mice were isolated and cultured by classical two-step digestion method and differential attachment method, and the purity of epidermal stem cells was identified as 90% by CD71 and CD49f flow cytometry.

[0041] Epidermal stem cells were seeded in 96-well plates, 10 wells in each group. When the original epidermal stem cells reached 70% confluency, administration was performed. The control group was added with 0.1% DMSO (dimethyl sulfoxide), and the GF-4592 group was added with working concentration 10 μM of FG-4592 (purity is 99.27%). After administration and treatment for 24 hours, CCK-8 cell proliferation detection was performed, and detection reagents were added according to the ratio of the instructions (in this embodiment, 90ul of medium and 10ul of CCK-8 we...

Embodiment 3

[0043] Example 3: Effect of FG-4592 on the proliferation of epidermal stem cells (2)

[0044] Experimental Design: The experiment was divided into control group and GF-4592 group. The epidermal stem cells of 1-day-old mice were isolated and cultured by classical two-step digestion method and differential adherence method, and the purity of epidermal stem cells was identified as 92% by CD71 and CD49f flow cytometry. Epidermal stem cells were inoculated on cell culture dishes and divided into two groups, with 2 dishes in each group as a repeat group. When the primary epidermal stem cells reached 70% confluency, administration was performed. The control group added 0.1% DMSO (dimethyl sulfoxide), and the GF-4592 group added FG-4592 (purity: 99.27%) with a working concentration of 10 μM. After 24 hours of drug treatment, the protein was extracted. Expression of HIF-1α and PCNA was detected by Western blot, see Figure 4 .

Abstract

The invention discloses application of FG-4592 to promotion of epidermal stem cell migration. On the basis of the FG-4592 structure and performances and specific administration route, dosage and administration frequency / time, epidermal stem cell migration can be effectively promoted by the FG-4592. According to researches, the FG-4592 is capable of promoting epidermal stem cell movement, remarkably increasing epidermal stem cell movement rate, promoting epidermal stem cell proliferation and improving epidermal stem cell proliferation activity, and a basis is provided for regulating epidermal stem cell migration to further regulate epidermal stem cell culture, epidermic grafting and wound healing. The FG-4592 can be applied to preparation of epidermal stem cell culture solution and auxiliary medicines for epidermic grafting and wound healing and brings good to patients suffering from burns, pressure sores, varicose ulcer of lower extremities and diabetic feet and patients with chronic and refractory wounds after skin flap transplantation and epidermic grafting, and a promising application and market prospect is achieved.

Description

technical field [0001] The present invention relates to the migration of epidermal stem cells, specifically the application of FG-4592 in promoting the migration of epidermal stem cells. Background technique [0002] Epidermal stem cells are cells with the potential to produce at least one highly differentiated progeny cell, located in the basal layer of the epidermis. Studies have shown that epidermal stem cells, as specific stem cells of skin tissue, are key cells for skin tissue generation, wound repair and reconstruction, and can participate in wound repair through proliferation, differentiation, and migration. Wound healing generally includes the following closely related and overlapping processes: coagulation phase, inflammatory response phase, epithelialization phase, and remodeling phase. Wound re-epithelialization is a key event that starts early and runs through the entire wound healing process. Plays an important role in restoring the integrity of the skin barrie...
