Abstract

The invention discloses a method for promoting aseptic germination of Dendrobium officinale and a culture medium used therein, and the application of the method. According to the invention, through research on phylembryogenesis and fruit maturity of the Dendrobium officinale, the aseptic germination conditions of the Dendrobium officinale are selected; a rapid propagation system is optimized; theproper seed age of sowing for aseptic germination of the Dendrobium officinale is determined; and Dendrobium officinale seeds with a proper seed age are sown into an improved solid culture medium to obtain protocorms with high germination rate and neat uniform germination state. The method provided by the invention enriches the reproductive biology development resources of the Dendrobium officinale and provides a theoretical basis for aseptic germination; through selection of the Dendrobium officinale seeds with a proper seed age as explants for tissue-culture rapid propagation, the method provided by the invention greatly shortens the propagation and breeding time of the Dendrobium officinale seeds and has more advantages in scale and industrialization development of the Dendrobium officinale. The protocorms obtained by culturing in the improved solid culture medium have high germination rate and neat and uniform germination state, and provide resources for the protection of wild resources through establishment of an aseptic germination system of the Dendrobium officinale and obtainment of excellent seedlings in production.
