Embodiment 1

[0054] In October 2015, the adult Pteris tricolor plants with good growth and development and no pests and diseases were selected in the wild, and the leaves with mature spores were cut off, and the spores without impurities were obtained by natural shedding and selection. Wrap the spores with double-layer lens-cleaning paper, soak in sterile water for 6 hours; then disinfect with 75% alcohol for 6 seconds, rinse with sterile water for 3 times, and rinse with 0.1% HgCl 2 The aqueous solution was sterilized for 6 minutes, and washed 5 times with sterile water repeatedly under aseptic conditions. Then pick up the lens-cleaning paper package with tweezers, cut the lens-cleaning paper package with scissors, and scatter the spores to the germination medium S1 (1 / 2MS+sucrose 20g / L+agar 5.6g / L, pH 5.8) by dragging the lens-cleaning paper )middle. Spore germination was first observed 15 days after inoculation, and the germinated spores were transferred to germination medium S2 (1 / 2MS...

Abstract

The invention provides a pteris tricolor tissue culture method and application thereof to preservation of germplasm resources, and relates to the technical field of plant tissue culture. The method takes a spore as an explant and comprises the following steps: 1) inoculating the explant in a germination culture medium for germination culturing to obtain a germinated seedling; 2) inoculating the germinated seedling into a multiplication inducing culture medium for performing multiplication inducing culture to obtain green globular bodies; 3) inoculating the green globular bodies into a differentiating-rooting culture medium for performing differentiating-rooting culture to obtain a rooted tissue cultured seedling. By adopting the pteris tricolor tissue culture method, a large quantity of rooted pteris tricolor cultured seedlings can be obtained after about 160 days; the multiplication rate is up to 1 to 10, the rooting rate is 90 percent or above, and the transplantation seedling rate is 80 percent or above. Moreover, the green globular bodies obtained through multiplication are preserved in vitro, so that the subculture period can be prolonged to 1.5 years or longer.

Description

technical field [0001] The invention belongs to the technical field of plant tissue culture, and in particular relates to a tissue culture method of Pteris tricolor and its application in germplasm preservation. Background technique [0002] Pteris tricolor is a plant of the genus Pteris in the family Pteridaceae. Its petioles and lobes are purple in color, and the center of the pinna has a wide band of white and rose red, so that the leaves are white, red and green as a whole. Tri-color stripes, hence the name. Pteris tricolor has beautiful plant shape and colorful leaves. It has high ornamental value and is a rare ornamental plant. At present, Pteris tricolor is only distributed in the subtropical area of southern Yunnan in China, and its habitat is narrow. [0003] Many species of Pteris genus are ornamental plants, which are widely used in gardening, plant landscaping, etc. Many species of this genus have had spore germination, green globule (GGB) induction and plant...
