Abstract

The invention discloses a sterile filter paper cultivation method for studying interaction between plant roots and environment. The method comprises following steps: S1. designing visual cultivation system, including a transparent cultivation container, a treatment medium, a triangular support and a growing medium; S2. sterilizing plant seeds and germinated, and after germination, transplanting the seedlings into a cultivation container to keep the plant growing naturally in the growth medium; S3. carrying out biotic or abiotic stress treatment on the germinated plant seeds, and tilting culture; S4. scanning roots by a camera system, a stereomicroscope and a scanner to perform two-dimensional in situ observation and image capture of roots. This method can directly perform 2-D visualization on the in-situ growth of plant roots without changing the growth environment of plant roots and its surroundings; combined with computer image analysis, in-situ observation and measurement of root response to the environment are carried out, and dynamic, in-situ and non-destructive root-environment interaction is observed and measured.
