Embodiment Construction

[0010] The present invention will be further described below in conjunction with specific embodiment, and concrete experimental procedure is as follows:

[0011] 1. Cloning of tomato transcription factor gene SolyWRKY41 gene

[0012] A pair of specific primers (forward: 5'-ATGGAGAAAGTTAAAAGTATGGA-3', reverse 5'-TTAAATGAAGAATTCTTCAATGTC-3') were designed using the cDNA of tomato 'Zheza 301' leaves as a template for amplification. The PCR reaction conditions were: pre-denaturation at 94°C for 5 min; denaturation at 94°C for 30 s, annealing at 54°C for 30 s, extension at 72°C for 1 min 10 s, a total of 35 cycles; extension at 72°C for 10 min. The target bands were recovered, ligated and transformed into Escherichia coli DH5α, and positive clones were picked and sent to Nanjing GenScript Biotechnology Co., Ltd. for sequencing.

[0013] 2. Subcellular localization of tomato transcription factor SolyWRKY41

[0014] Through nucleotide sequence determination, the sequence of tomato ...

Abstract

WRKY transcription factor family widely exists in plants. In accordance with the quantity of WRKY structural domains and the characteristics of a zinc finger structure, the WRKY transcription factor family can be divided into three sub-families (I, II and III). An WRKY transcription factor gene Soly WRKY41 is analyzed and identified from a tomato variety which is infected by tomato yellow leaf curl virus. The transcription factor has one WRKY structural domain and a zinc finger structure of C2HC, belonging to members of the WRKY III sub-family. Based upon subcellular localization analysis, itis indicated that the SolyWRKY41 transcription factor belongs to nucleoprotein. A virus-induced gene silencing result shows that the SolyWRKY41 gene can implement negative regulation of the tomato yellow leaf curl virus. Through functional researches of the SolyWRKY41 transcription factor, the molecular mechanism of tomatoes in response to the tomato yellow leaf curl virus is enriched; and the SolyWRKY41 transcription factor is applicable to tobacco disease-resistance breeding.

Description

technical field [0001] The invention belongs to the field of plant genetic engineering and relates to the application of a WRKY III type transcription factor in plants. The present invention identifies a WRKY III transcription factor gene SolyWRKY41 from tomato that responds to tomato yellow leaf curl virus, and the gene is helpful for the research on the resistance mechanism of tomato responding to tomato yellow leaf curl virus. Background technique [0002] Tomato (Solanum lycopersicum L.) belongs to Solanaceae (Solanaceae), tomato genus (Solanum), rich in nutrients such as sugar, dietary fiber, minerals, amino acids and vitamins, and plays an important role in balancing people's diet. Tomatoes can not only lower blood fat and blood pressure, anti-fungal, anti-inflammatory, but also prevent gastric cancer, especially lycopene has anti-oxidation effect, and is widely used in cosmetics and pharmaceuticals. In recent years, the cultivation area of tomato has been continuou...
