Problems solved by technology

Although this identification method can be used as an indicator substance for identifying natural leather, it can detect pyrolysis products derived from Hyp equivalent to dipeptides that are not affected by decomposition inhibition caused by pollution, but these pyrolysis products will not be caused by animals. Significant differences occur between different species, making it difficult to identify animal species

Embodiment 1

[0066] In this example, an example of identification of natural leather of 6 kinds of animals including cattle, horses, pigs, sheep, goats, and deer will be described.

[0067]First, the tissue of the skin of an animal species of natural leather and the collagen of the main fiber will be described. Animal skin consists of the epidermis, dermis and underlying subcutaneous connective tissue. The epidermis is composed of 4 layers, including the cuticle and the granular layer, and the layers that are not used by leather are removed. In addition, the elastin fibers in the papillary layer and subcutaneous tissue are almost completely removed during mechanical manipulation or drug handling during manufacturing. In addition, keratin, which makes up hair, is useless when making leather except for fur, and is decomposed and removed by lime treatment. Therefore, the fibers that make up leather are mostly collagen fibers. The dermis is divided into the papillary layer (silver layer) an...

Embodiment 2

[0110] In this example, an example of identification of animal species of fur such as foxes, raccoons, and minks will be described.

[0111] About the process flow of the identification method of the animal species of the fur of fox, raccoon, mink etc., also same as embodiment 1, follow figure 1 process.

[0112] First, keratin is extracted from the fur of fox, raccoon, mink, etc. (step S101). The extracted keratin is decomposed with keratinase (step S102). As the keratin-decomposing enzyme, trypsin can be used.

[0113]Next, in the same manner as in Example 1, the amino acid sequences of the peptides are analyzed (step S103), the detected peptides that are identical to all identified animal species are removed (step S104), and the peptides detected from the animal species are selected. specific peptides (step S105). Then, use the selected peptides to identify the animal species of the sample (step S106). 【Other Embodiments】

Abstract

[Problem] To provide an objective method and a device for distinguishing animal species by identifying animal species through peptide analysis. [Solution] This method is provided with: a step for, byusing a proteolytic enzyme (protease), cleaving, into peptide fragments, a group of animal fibrous proteins (collagen, etc.) extracted from a distinguishing-subject sample, and analyzing the peptide fragments through amino acid sequencing; a step for removing, from all the analyzed peptide fragments, peptide fragments commonly detected in all animal species to be distinguished, and selecting peptide fragments that are specifically detected in at least one animal species; and a step for distinguishing the animal species of the sample by using the selected peptide fragments. Among the selected peptide fragments, a peptide fragment detected specifically in one particular animal species is used first for distinguishing the animal species of the sample, and, with respect to a sample with whichdistinguishing was unsuccessful, a peptide fragment detected specifically in at least two animal species is used next for distinguishing the animal species.

Description

technical field [0001] The present invention relates to a technology related to an objective identification method of animal species, especially a method for identifying leather species. Background technique [0002] According to the Household Goods Quality Indication Law, there are regulations on the quality indication of miscellaneous industrial goods for leather bags, clothes, gloves, and furniture. Among them, for clothing that uses leather or synthetic leather as all or part of the product, identification of the six types of animals (cattle, pigs, sheep, goats, horses, and deer) that are indicated is required. [0003] Recently, in order to express the quality correctly, more and more products are required to have accurate animal identification in the distribution stage. However, there is currently no legal identification method. Regarding the expression of these miscellaneous industrial products, the methods usually used for identification in examinations or inspecti...
