Problems solved by technology

After using this method to treat sesame seeds, the disease resistance level of sesame tends to be stable 4 weeks after inoculation (that is, the stage of two pairs of true leaves of sesame cultivars), which is not conducive to further observation of sesame diseases; for some strong pathogenic FOS strains, large Most sesame materials will all die at the cotyledon stage, resulting in incomplete symptoms of wilt disease, and to a certain extent, affecting the observation of the pathogenic process of pathogenic bacteria and the monitoring of sesame disease resistance and response process

Embodiment 1

[0053] In this example, taking Yuzhi 11 and Y11-M9956 as examples, the fusarium wilt resistance of these two varieties (lines) was preliminarily identified. The specific process is briefly introduced as follows.

[0054] (1) Prepare the sesame plants

[0055] Pick 100 plump seeds of two sesame varieties (lines) Yuzhi 11 and Y11-M9956 respectively, sterilize them, and then shake them at 28°C and 120rpm until the seeds are white;

[0056] The specific procedure of the sterilization treatment is as follows: first treat with 70% alcohol for 30 seconds, then treat with 3% sodium hypochlorite for 10 minutes, and finally rinse with sterile water for 3 to 5 times;

[0057] Sow the Lubai seeds in a paper cup filled with sterile vermiculite, and 10 seeds can be sown in each paper cup;

[0058] Irrigate the improved Hoagland nutrient solution once every 3 days, cultivate in an artificial climate incubator at a temperature of 25-28°C, a relative humidity of 60-80%, and alternate light an...

Embodiment 2

[0096] In this example, 5 FOS strains of Fusarium oxysporum (HSFO15050, HSFO15098, HSFO15099, HSFO15100, and HSFO15110) were taken as examples to evaluate the pathogenicity of sesame. The specific experimental process is briefly introduced as follows .

[0097] (1) Prepare the sesame plants

[0098] The seeds of two sesame varieties (lines) Yuzhi 11 and Y11-M9956 were selected for pathogenicity evaluation, and the specific cultivation and planting process of sesame materials was the same as in Example 1.

[0099] (2) Preparation of conidia suspension for dipping

[0100] For different Fusarium oxysporum sesame-specific (FOS) strains (HSFO15050, HSFO15098, HSFO15099, HSFO15100 and HSFO15110), the preparation process of the conidia suspension was the same as in Example 1. The number of bacteria in the spore suspension was adjusted to 1×10 6 cfu / mL.

[0101] (3) root soaking inoculation

[0102] The sesame plants of Yuzhi No. 11 and Y11-M9956 to be tested were respectively i...

Abstract

The invention specifically relates to a method for identifying and evaluating the level of wilt resistance of sesame, belonging to the field of timely evaluation of the disease resistance of crop varieties. The method comprises the following steps: preparation of sesame plants; preparation of a conidium suspension used for dip dyeing; dipping of roots and inoculation of bacteria; identification and evaluation of the wilt resistance of a sesame variety; etc. The method provided by the invention can realize inoculation of sesame plants in different growth stages (from a stage of two pairs of true leaves to a squaring stage) on the basis of a mature root-dipping infection method. Meanwhile, since the concentration of a wilt strain for inoculation is easy to adjust, so detailed analysis can be carried out on both the pathogen infection process and the response process of sesame seedlings, the disease resistance of different sesame germplasms and the pathogenicity of different strains can be accurately identified and evaluated, a technical base is provided for effective control of sesame wilt, and reference is provided for control of other crop diseases. The method provided by the invention has good repeatability, stable and reliable identification results and good application value in scientific research.

Description

technical field [0001] The invention belongs to the field of crop variety disease resistance evaluation and in particular relates to a fusarium wilt resistance identification and evaluation method for sesame. Background technique [0002] Sesame is an important characteristic oil crop in my country. Sesame Fusarium Wilt is mainly caused by Fusarium oxysporum Sesame Specialization ( Fusarium oxysporum f. sp. sesami , FOS) is also a worldwide sesame disease. Together with stem point blight, it is called the two major fungal diseases in sesame production. The annual incidence of sesame wilt is about 15%, and it can reach more than 30% in severe cases, which has a great impact on yield and quality. [0003] In recent years, in order to detect the pathogenicity of the sesame-specific type of Fusarium oxysporum and evaluate the resistance level of sesame germplasm to Fusarium wilt, the Sesame Research Center of Henan Academy of Agricultural Sciences used the method of seed inoc...
