Problems solved by technology

At present, the amplification time of traditional STR detection kits is about 3 to 4 hours. If the amplification time can be controlled within 1.5 hours, the detection efficiency and kit performance will be greatly improved.

Embodiment 1

[0058] Blood was provided by volunteers. Template DNA was extracted by chelex-100 method. The amplification reaction was carried out on the ABI9700 thermal cycler, the electrophoresis and detection were carried out on the ABI 31XL genetic analyzer, and the data analysis was carried out using GeneMapper IDv3.2 software. At the same time, the samples were detected by the Goldeneye 20A kit of Genepoint Cognitive Co., Ltd., and the operation method was carried out according to the kit instructions, and the results were used as a control. The reagents and materials used in the present invention, such as allelic ladder (ladder), are conventional materials commonly used by those skilled in the art.

Abstract

The invention relates to a composite amplification method and kit for STR loci. The composite amplification method and kit are used for amplifying 22 STR loci and one sex locus, wherein the 22 STR loci are composed of vWA, D21S11, D18S51, D5S818, D7S820, D13S317, D16S539, FGA, D2S1338, D19S433, D6S1043, D12S391, D8S1179, D3S1358, D1S1656, CSF1PO, Penta D, Penta E, TH01, TPOX, D10S1248 and DYS391, and the sex locus is Amelogenin. Amplimers used in the composite amplification method and kit comprise sequences as shown in SEQ ID No. 1 to 46. The composite amplification method and kit provided by the invention can acquire information about the 22 STR loci with good polymorphism and high individual discrimination power and about the one sex locus through simultaneous amplification at a time; and a composite amplification system composed of the loci has good balance, high sensitivity and high specificity, is accurate in typing results and can totally meet demands of judicial expertise.

Description

technical field [0001] The invention relates to the technical field of fusion proteins, relates to the detection of polymorphic genetic marker genes in the human genome, and in particular to a method for compound amplification of 22 STR (short tandem repeat) loci and 1 sex locus and its Reagent test kit. Background technique [0002] Short tandem repeats (STR for short), also known as microsatellites or simple sequence repeats (SSR), are a type of DNA tandem repeats widely present in eukaryotic genomes, with a core sequence of 2-6 base repeat units. The number of STR loci is large and widely distributed, accounting for about 3% of the entire genome (International Human Genome Sequencing Consortium, 2001), and the polymorphism is high. Species differences follow the Mendelian law of inheritance in the genetic process. Therefore, STR amplification detection technology is widely used in forensic individual identification, kinship identification and population genetics researc...
