Problems solved by technology

However, this paradigm also has major flaws: 1. There are various causes of cancer, and the subdivision by pathogenic gene is too fine.
Second, the research and development of new drugs has a large workload, a long cycle, and slow returns
Third, cancer is heterogeneous. Targeting and killing specific clones of cancer cells often leads to the expansion of other clones, resulting in recurrence and drug resistance

Embodiment 1

[0039] Example 1: Lovastatin selectively induces apoptosis in multiple myeloma cell lines--sample matching based on biological pathways related to the drug's mode of action

[0040] In this example, the transcriptome expression profile data (PMID: 20360469) of 4 multiple myeloma cell lines before and after lovastatin treatment published by Clendening JW et al. were collected online. Among them, H929 and KMS11 were lovastatin-sensitive cell lines, and apoptosis occurred 12 hours after adding lovastatin. LP1 and SKMM1 are lovastatin-insensitive cell lines, which do not undergo apoptosis after adding lovastatin.

[0041] In this example, the multiple myeloma cell line H929 was used as a cell line with a known phenotype after drug treatment (that is, the model responding to lovastatin in the drug response model library), and the other three groups of multiple myeloma cell lines KMS11, LP1 and SKMM1 were used as cell lines with unknown post-treatment phenotypes (ie new cases).

Embodiment 2

[0054] Example 2: Lovastatin selectively induces apoptosis of multiple myeloma cell lines--sample matching based on expression profile data before and after model drug response in the model library

[0055] The source of data in this example is the same as that in Example 1, using the transcriptome expression profile data of four multiple myeloma cell lines before and after lovastatin treatment published by Clendening JW et al. This example mainly utilizes the characteristic signal molecules of H929 in response to drugs before and after administration of lovastatin, and provides another way of matching with the drug response model.

[0056] In this example, the difference in expression profile of H929 before and after administration of lovastatin was compared, and the 100 genes with the most significant difference in expression profile were also ranked according to the significance of the difference as the characteristic signal molecules of the characteristic class for drug res...

Abstract

The invention discloses a cancer individuation intervention strategy selection method based on molecular physiome. The method comprises the following steps that: establishing an intervention response model library; when a new case appears, comparing the new case with the pre-administration proteomics difference of a model; and analyzing biological approaches / physiological function differences and the like which may be caused by the difference. A sample data type on which the method depends has high dependency with a physiological level phenotype. Proteomics data related to cell physiology and functions including protein and the like can be transcribed and subjected to epigenetic inheritance. The method is more sensitive to the intervention response effect of diseases, can provide rich indexes to observe molecular level physiological changes caused by diseases, and is suitable for clinic transformation and practical popularization.

Description

technical field [0001] The invention belongs to the field of personalized medication in the medical and health industry, and specifically relates to a method for selecting an individualized cancer intervention strategy based on individual molecular physiological omics data. Background technique [0002] The structure of the modern medical system is based on a limited observation index system. The essence of evidence-based medicine is to classify patients according to the diagnostic indicators obtained from various examinations so as to prescribe the right medicine. This kind of research based on objective and real observation indicators rather than "imagination" has always been the foundation of medicine, and this point will not change in the past, present and future. [0003] However, the limited index system of modern medicine makes the existing disease diagnosis and classification framework relatively rough. Classifying patients using disease concept definitions under t...
