Problems solved by technology

Moreover, whether it is traditional electron microscope observation, serological detection method or RT-PCR method, they all require the tester to have higher experimental conditions and equipment, and these are exactly what beekeeping farmers do not have.

[0022] 1. Materials

[0023] The samples of Chinese bees infected with cystic larval disease virus of Apis mellifera were collected and preserved by our laboratory. The mouse monoclonal antibody against the vp3 capsid protein of Apis cystic larvae disease virus was entrusted to Shanghai Wenyuange Biotechnology Co., Ltd., and two different hybridoma cell lines were obtained after screening and preserved by our laboratory. Goat anti-mouse IgG antibody, trisodium citrate, chloroauric acid (HAuCl4 4H2O) and bovine serum albumin (BSA) were purchased from Sigma Company; nitrocellulose membrane, absorbent paper, PVC bottom plate, glass fiber membrane were purchased from Shanghai Jie One Biotech Co., Ltd. All other reagents were of domestic analytical grade.

Abstract

The invention discloses an immunity colloidal gold test strip for quickly detecting Chinese bee sac brood virus. The immunity colloidal gold test strip comprises a PVC (Polyvinyl Chloride) base plate, wherein a sample pad, a colloidal gold pad, a nitrocellulose membrane and a water absorption pad are sequentially fixed on the PVC base plate from left to right; a detection line and a quality control line mutually separated are arranged on the nitrocellulose membrane; the colloidal gold pad is marked with an anti-mouse infectious bursal disease virus capsid protein vp3 monoclonal antibody; the detection line on the nitrocellulose membrane is coated with another anti-mouse vp3 monoclonal antibody; the quality control line on the nitrocellulose membrane is coated with goat-anti-mouse IgG. The colloidal gold test strip is used for detecting the Chinese bee sac brood virus, and is high in detection specificity, simple, convenient and fast to operate, intuitive in result, and convenient for popularization and application.

Description

technical field [0001] The invention belongs to the technical field of virus epidemic disease diagnosis, and relates to a colloidal gold test strip for immunity to sac larvae disease virus in honey bees. Background technique [0002] Chinese sacbrood disease is caused by Chinese sacbrood virus (CSBV), which is a serious virus disease to Chinese bees. The symptoms are mainly seen in the larvae of the bee. The virus replicates in large quantities in the larvae, resulting in the death of the larvae quickly, and the subcutaneous exudate of the worm body increases. The bee sac larvae disease has a rapid onset, wide spread and high mortality. If the bee colony is sick, it will affect the reproduction and collection of the bee colony, and if it is severe, it will cause the entire bee colony to be wiped out. There is no specific drug treatment for this disease at present, and it is a major problem that needs to be solved urgently in the modern breeding of Chinese bees. In my count...
