Problems solved by technology

Molecular dynamics simulation provides convenience for the rapid screening of highly efficient skin light-transparent reagents, which can not only avoid blindness and time-consuming experimental screening methods, but also have good consistency between theoretical predictions and experimental results, such as academic papers: W.Feng* , R.Shi*, N.Ma, Daria.K.Tuchina, Valery V.Tuchin, and D.Zhu, "Skin optical clearing potential of carbohydrates" Journal of Biomedical Optics, 21(8): 081207(2016) : Theoretical simulations and experiments have shown that sucrose has a better light-transparency effect than fructose. However, pure sugar reagents are hindered by the skin stratum corneum and dense epidermis, and their role in improving high-resolution in vivo imaging of skin vascular structure and function Limited capabilities, such as academic papers: J.Wang, N.Ma, R.Shi, Y.Zhang, T.Yu, and D.Zhu, "Sugar-induced skin optical clearing: Frommolecular dynamics simulation to experimental demonstration," IEEE Journal ofSelected Topics in Quantum Electronics, 20(2): 7101007(2014). Affected by it, the in vivo application of sucrose in skin light transparency and its ability to improve in vivo optical imaging research are not optimistic

Abstract

The invention provides a skin tissue optical clearing solution, and a preparation method and applications thereof. According to the preparation method, the skin tissue optical clearing solution comprises, by volume percent, 40 to 55% of an aqueous solution of a monosaccharide and a derivate of the monosaccharide or a disaccharide and a derivative of the disaccharide, 40 to 50% of an alcohol polymer, 5 to 10% of a penetration enhancer, and a dissolution enhancer which can be dissolved in water and organic solvents, wherein the volume of the dissolution enhancer is 3 to 5% of the total volume of the former three ingredients. When the skin tissue optical clearing solution is applied to skin tissues, uniformization of the refractive index in the skin tissues is realized, tissue scattering is reduced, the skin tissues become transparent to light, imaging depth is increased, and a novel method used for acquiring skin capillaries structures, and blood flow and blood oxygen distribution information is provided.

Description

technical field [0001] The invention belongs to the technical field of biomedical optical imaging, and in particular relates to a skin tissue light transparent solution and a preparation method and application thereof. Background technique [0002] Skin is an important target of in vivo optical imaging research, which is of great significance in the research of peripheral vascular disease, diabetes, tumor, microvascular injury and remodeling, immunology and other fields. The use of optical imaging technology can not only monitor the skin blood vessels, blood flow and blood oxygen metabolism level, but also track the movement behavior of inflammatory cells during the immune response at the single cell level. However, the turbid nature of the skin limits the depth of light penetration in tissue, making these techniques only able to image superficial tissues with low image contrast and resolution. The proposal of biological tissue light-transparency technology provides a new i...
