Problems solved by technology

my country's fir tissue culture research has developed rapidly in the past ten years, and fir tissue culture production lines have been established in Fujian, Guangdong and other places. However, the existing technology generally has problems such as high production costs and low seedling cultivation results, and there is no efficient and economical tissue culture method to support it. The establishment of high-quality seedling production bases, coupled with the insufficient output of existing seed orchards, has led to a shortage of seedlings of Chinese fir plantations, and the genetic quality of the species used for production is uneven. How to economically and quickly breed a new generation of fine species is imminent
Only relying on the existing Chinese fir tissue culture seedling rooting and seedling hardening technology to carry out seedling breeding cycle is long, transplanting survival rate is low, there is no relevant report on the method of shortening the Chinese fir tissue culture seedling rooting and hardening time

Embodiment 1

[0025] 6 Chinese fir clones were cultivated in the Guangdong Academy of Forestry Sciences, a total of 6,000 seedlings were rooted, and 5,000 seedlings were transplanted after hardening. The Chinese fir tissue culture seedling rooting culture that present embodiment adopts and the method for hardening seedling is specific as follows:

[0026] (1) Selection of tissue cultured shoots for rooting culture: On September 15, 2015, among the Chinese fir tissue cultured shoots for multiplication culture, the shoots with strong growth, dark green leaves and a length of more than 3 cm were selected for rooting to cultivate;

[0027] (2) Rooting inoculation: on the ultra-clean workbench, cut the part of the top tip of the tissue cultured shoots used for rooting culture that is 2.5 cm to 3.0 cm down, and insert it into the glass culture bottle that has prepared the rooting medium in time 12 to 15 seedlings were inoculated in each bottle, and a plastic sealing cap was added. The volume of ...

Abstract

The invention relates to a rooting culture and hardening seedling method of a cunninghamia lanceolata tissue-cultured seedling. The method comprises steps of selection of tissue-cultured bud seedlings, rooting and inoculation, rooting culture, rooting and cap-sealed hardening seedling, and uncapped hardening seedling. The method combines rooting with hardening seedling, short-period rooting culture is carried out in a culture room, and rooting and cap-sealed hardening seedling and uncapped hardening seedling culture are carried out, so that the tissue-cultured seedling is subjected to rooting and hardening seedling at the same time. Time for rooting and hardening seedling of the cunninghamia lanceolata tissue-cultured seedlings is obviously shortened. Compared with a conventional method, the method allows the seedling culture period to be obviously shortened, provides suitable root length, and obviously improves the seedling culture efficiency. Energy and expense saving is achieved through natural temperature control and light control. The problems of high cost and low survival rate of transplanting in a production process of a cunninghamia lanceolata tissue-cultured seedling are solved. The method is beneficial for establishing an excellent asexual seedling culture base for cunninghamia lanceolata and provides superior seedlings for forestry production.

Description

technical field [0001] The invention relates to the field of agricultural technology, in particular to a method for rooting and hardening Chinese fir tissue culture seedlings. Background technique [0002] Chinese fir (Cunninghamia Lanceolata) belongs to the Taxodiaceae family. Chinese fir is an evergreen tree species with fast growth and good material quality. It is one of the important fast-growing commercial timber tree species in southern my country. Plant tissue culture is an important method for rapid propagation of new plant varieties. While maintaining the excellent genetic characteristics of varieties, it can breed high-quality seedlings in a short period of time for production and application, which greatly promotes the process of improving industrial timber forests. my country's fir tissue culture research has developed rapidly in the past ten years, and fir tissue culture production lines have been established in Fujian, Guangdong and other places. However, the e...
