[0008] Hemolysis test

[0009] The purified pneumolysin ply was serially diluted twice in a 96-well plate, and 10 7 Sheep red blood cells were incubated at 37°C for 10 min and then centrifuged. The highest dilution factor that lyses red blood cells is regarded as the hemolysis titre (Hemolysis titre)

[0010] The hemolytic titers of hemolysin ply without adding abiflavone and adding different concentrations of abiflavone are as follows (Table 1):

[0013] Protective Test of Injured Human Lung Epithelial Cells (A549)

[0014] A549 cells were cultured in DEMED medium (containing 10% fetal bovine serum) at 37°C with 5% CO 2 Cultured to 1.5 x 10 4 Cells / well were plated in a 96-well cell culture plate, 100 μl per well. After the cells adhered to the wall, add 0.5 μl of purified pneumolysin protein, and add different concentrations of abiflavone, and pl...

Abstract

The invention relates to application of amentoflavone in research and development of medicines for treating pneumonia. Rabbit erythrocyte haemolysis tests, human pulmonary epithelial cell (A549) injury protection tests and a mouse streptococcus pneumonia model prove that the amentoflavone has a treatment effect on infection of streptococcus pneumonia. Compared with treatment with antibiotics, the amentoflavone for treatment has the characteristics of no drug resistance and high protection performance.

Description

technical field [0001] The invention relates to the application of the biflavone of Suicha spica in the research and development of medicines for treating pneumonia, and belongs to the field of medicine. Background technique [0002] Amentoflavone (Amentoflavone) is a kind of polyphenolic compound, which widely exists in Selaginella in nature. The biological activities of anti-inflammation, anti-tumor, anti-radiation, scavenging free radicals and neuroprotection of biflavonoids have attracted more and more people's attention. But so far, there are no reports on the use of biflavonoids in the treatment of bacterial pneumonia at home and abroad. [0003] Bacterial pneumonia is a disease caused by infections such as Streptococcus pneumoniae, Haemophilus influenzae, Staphylococcus aureus, Streptococcus pyogenes, Legionella, etc. The mortality rate of this disease is extremely high in children, the elderly and immunocompromised patients. In recent years, the extensive use of an...
