Embodiment 1

[0031] Example 1: Screening characterization of differential metabolites in plasma between subjects with normal coronary arteries and patients with acute coronary syndrome

[0032] 1. Objects and methods

[0033] 1. The source of the specimen

[0034] After obtaining the consent of the patients, the peripheral venous blood plasma of 460 patients with acute coronary syndrome and 350 subjects with normal coronary arteries in Jiangsu Provincial People's Hospital from September 2010 to June 2015 were collected. Confirmed by angiography. Patients with normal coronary arteries were age and sex matched with patients with acute coronary syndrome. All patients with acute coronary syndrome and normal coronary arteries have normal heart, lung, liver, kidney and hematopoietic functions.

[0035] The time of blood collection was in the morning on an empty stomach.

[0036] 2. Main reagents

[0037] Acetonitrile and formic acid (UPLC pure) were purchased from ROE Company in the United ...

Embodiment 2

[0058] Example 2: Construction of ROC curve to verify the ability of 7 differential metabolites for diagnosis of acute coronary syndrome

[0059] Receiver operating curve (ROC) method was used for verification, and the ability to diagnose coronary heart disease was judged by the expression levels of differential metabolites in the plasma of patients with acute coronary syndrome and normal coronary arteries. The results showed that phosphorylcholine, palmitoylethanolamine, lysophosphatidylcholine (16:0), phosphatidylceramide (d18:1 / 8:0), lysophosphatidylcholine (18:2), lysophosphatidylcholine The seven differential metabolites of choline (18:1) and phosphatidylinositol (18:3 / 0:0) have a strong ability to diagnose patients with acute coronary syndrome and normal coronary arteries, and the ROC curve The area under the area (AUC) is greater than 0.7, which has clinical diagnostic significance; when combined for diagnosis, with the increase of the number of combinations, the AUC fu...

Embodiment 3

[0069] Example 3: Application in Screening for the Treatment or Relief of Drugs for Acute Coronary Syndrome

[0070] The plasma of 190 patients with acute coronary syndrome before and after drug treatment was detected, and the results showed that: with the increase of the course of drug treatment, the patient's condition was gradually relieved, and the levels of 7 metabolic markers provided by the present invention in plasma The level tends to the level of healthy people gradually, and there is no significant difference between the plasma level of healthy people after complete cure (P>0.05). This result is consistent with the detection results of coronary angiography. Therefore, the metabolic markers provided by the present invention can be used to screen drugs for treating or alleviating acute coronary syndrome.

Abstract

The invention discloses a metabolism marker for diagnosis of the acute coronary syndrome. The metabolism marker comprises one or more of phosphorylcholine, palmitoylethanolamine, lysophosphatidyl choline (16:0), phosphatidyl ceramide (d18:1 / 8:0), lysophosphatidyl choline (18:2), lysophosphatidyl choline (18:1) and phosphatidylinositol (18:3 / 0:0). When a single metabolism marker is used for diagnosis and distinguishing of acute coronary syndrome patients and coronary artery normal people, ROC area under the curve (AUC) is larger than 0.7 all the time, and clinical diagnosis significance is high; when several metabolism markers are combined to be used for diagnosis, AUC is further increased as the combination number increases and reaches the highest value 0.987 when all the seven metabolism markers are combined, and sensitivity and specificity are 99.1% and 98.9% respectively under the optimum cutoff value. The metabolism marker can be used for accurate diagnosis of the acute coronary syndrome and has high accuracy, sensitivity and specificity.

Description

technical field [0001] The invention belongs to the field of biochemistry, and relates to metabolic markers for diagnosis and classification of coronary heart disease, in particular to a group of metabolic markers for diagnosing acute coronary syndrome. Background technique [0002] Coronary heart disease, also known as ischemic heart disease, involves atherosclerosis in the arteries supplying myocardial blood, that is, coronary atherosclerotic lesions cause vascular lumen narrowing or plaque formation or even rupture and complete blockage, resulting in myocardial ischemia and hypoxia. Or necrosis leads to cardiac ischemic disease, which leads to a series of serious cardiovascular events such as clinical angina pectoris and myocardial infarction. Coronary heart disease is the main killer of human health. It has the characteristics of high incidence rate, high disability rate, high recurrence rate, high mortality rate, and many complications. It has become a major disease tha...
