Abstract
The invention discloses a method for regulating and controlling peltate yam rhizome triploid plants to root. The method is characterized by comprising the specific steps of selecting peltate yam rhizome triploid regeneration buds which grow healthily, and cutting the regeneration buds into stems with buds, which are 2-3cm long, on a clean bench; inoculating the stems into a culture medium, wherein the culture medium comprises a 1 / 2MS, 0.3mg / L of NAA (Naphthalene Acetic Acid), 0.24-0.96mg / L of ecdysterone, 30000mg / L of sucrose and 7500mg / L of agar powder, and the pH value of the culture medium is 5.8-6.0; and culturing under aseptic conditions, wherein the culture conditions are as follows: the temperature is 22-24 DEG C, the stems are cultured at a bright place for 16 hours and at a dark place for 8 hours, the relative humidity is 80%-90% and the illumination intensity is 2000-3000Lx. The method has a remarkable facilitation effect on the rooting of the peltate yam rhizome triploid plants; and particularly, the rooting starting time can be shortened, the average root length and the average rooting quantity are remarkably increased, and the transplanting survival rate is also remarkably improved.
