In a tube, tip, or other container, the collected sample is combined with lysis buffer for lysis of cells or particles, and stabilization and binding of nucleic acids. The lysis buffer may comprise guanidine thiocyanate, isopropanol, triton X-100, pH 7 MOPS buffer, and carrier RNA. The released nucleic acids bind to the inner surface of the container (or to a solid phase contained in the container, such as beads), and unbound material (eg, salts, proteins, cell debris, and other debris) is removed. Samples were then washed by adding and removing wash buffer. The wash buffer may comprise MOPS buffer at pH 7, salt (eg, NaCl), and ethanol. Washing can be performed once, or can be repeated to increase the purity of subsequent eluates. After the wash buffer is removed, an elution buffer is added to release bound nucleic acids.
