Abstract

The invention relates to an electrochemical biosensor as well as a preparation method and application thereof. The preparation method comprises the following steps: (1) preparing drugs for synthesizing a polymer membrane into a solution with a certain concentration for standby; (2) dissolving a DNA sequence in a Tris-HCl buffer solution for standby; (3) polishing a gold electrode, and then conducting ultrasonic cleaning; (4) taking the cleaned bare gold electrode as a working electrode, taking the polymer solution with certain concentrate of template molecules as an electrolyte solution, and utilizing an electric polymerization process to synthesize the polymer modified gold electrode; (5) soaking and cleaning the obtained polymer modified electrode, and removing the template molecules in the polymer to obtain the molecularly imprinted membrane modified gold electrode; (6) obtaining the electrochemical biosensor established based on the molecularly imprinted polymer membrane. According to the invention, the advantages of the electrochemical biosensor and the template molecule technology are combined, and the synthetic method is simple, and low in energy consumption and cost; the template molecules can be released through the utilization of the specific binding function between the target analyte and the template molecules namely folic acid or the dissection function of the excision enzyme III, so that purpose that the folic acid receptor and mercury ions are respectively detected can be achieved.
