Embodiment 1

[0030] Embodiment 1, the method for optimizing the polysaccharide extraction process of Dendrobium officinale by response surface methodology:

[0031] 1. Sample preparation

[0032] 1. Sample preparation

[0033] Dendrobium officinale is dried to moisture content≤8% (weight content), pulverized, passed through a 100-mesh sieve, and used as a sample.

[0034] Weigh 0.50 g of the sample with a balance, add an organic solvent (ethanol, 10 ml) to carry out decolorization pretreatment, then centrifuge to remove the organic layer, add deionized water according to the material ratio, and extract at different temperatures for a certain period of time, the extracted solution Centrifuge (rotating speed 10000, 10 minutes, 15° C.), recover the supernatant, use deionization to make up to the same volume, and use the modified phenol-sulfuric acid method to quantitatively measure the content of polysaccharides. The yield was calculated with the formula: yield (%)=[weight of polysaccharide...

Abstract

The invention discloses an extracting process of dendrobium candidum polysaccharides. The process comprises the following steps of: mixing dendrobium candidum and water according to a ratio of liquid to material of 51-52 ml of water per 1 g of dendrobium candidum, and extracting at 92.5 DEG C to 93 DEG C for 2.5-2.6 hours to obtain dendrobium candidum polysaccharide extract. The invention further provides a method for optimizing the extracting process of the dendrobium candidum polysaccharides by a response surface method, and the method comprises the following steps: setting three variables: a ratio of liquid to material of the dendrobium candidum to the water, extracting temperature and extracting time; and optimizing by utilizing a design expert software according to a Box-Behnken design principle. By adopting the extracting process provided by the invention, the yield of the dendrobium candidum polysaccharides can be improved, the consumption of water resources is reduced, and the generation and emission of sewage in a production process can be decreased.

Description

technical field [0001] The invention relates to a method for optimizing the extraction conditions of dendrobium officinale polysaccharides, in particular to a method for optimizing the extraction conditions of dendrobium officinale polysaccharides by using the response surface method. Background technique [0002] Dendrobium officinale is a perennial epiphytic herb of Orchidaceae. Dendrobium candidum has unique medicinal value, and its stem is used as medicine. The traditional Chinese medicine is called Dendrobium, which belongs to the yin-tonifying medicine in the tonic medicine "Chinese Pharmacopoeia". The tender stems of Dendrobium officinale are twisted into a spiral or spring shape, and dried in the sun. The commercial product is called Fengdou. Dendrobium officinale has become an important traditional Chinese medicine species in Zhejiang Province. Dendrobium candidum is rich in polysaccharides, which can enhance immune function. Preliminary experiments showed that D...
