Abstract

The invention provides a method for removing endotrophic bacteria in a tissue culture system of a nandina with red leaves, which belongs to the technical field of plant cultivation. The method is characterized by comprising the following steps of: (1) selecting tissue culture seedlings with endotrophic bacterium infection in seedling bases in a culture medium from the tissue culture seedlings of the nandina with red leaves in a proliferated state; (2) taking out the tissue culture seedlings with endotrophic bacteria on an ultraclean workbench, cutting off apexes and peeling off stem apexes; (3) inoculating the peeled stem apexes into an inducing differentiation culture medium added with bacteriocidal substances; an (4) cutting off adventitious buds differentiated from the stem apexes after the stem apexes are cultured for a section of time, transferring into a proliferation culture medium to be cultured until the tissue culture seedlings grow normally and no endotrophic bacterium pollution is caused. By using the removing method, the removing rate of the endotrophic bacteria reaches 90%, and the obtained nandina with red leaves has excellent quality and broad application prospect.
