Problems solved by technology

[0007] Although TTV may play a role in the development of diseases such as PMWS, it is difficult to rule out the possibility that the disease material itself contains other pathogens because the above research used the disease material of pigs infected with TTV1

Embodiment Construction

[0017] Considering that there is a high degree of variation in the porcine TTV1 nucleotide sequence, the present invention is based on three currently published porcine TTV1 genome sequences (Sd-TTV31, AB076001; 1p, AY823990; swSTHY-TT27, GQ120664), first by amplifying the virus The nucleotide sequence of the specific region of the non-coding region is relatively conserved, and then a pair of reverse primers are designed according to the obtained nucleotide sequence to amplify the nucleotide sequence of the complete genome of the virus.

[0018] First, extract DNA from porcine serum samples according to the conventional phenol-chloroform method, that is, take 300 μL of serum, add an equal volume of lysis buffer and 10 μL of proteinase K (10 mg / mL), digest at 50 °C for 2 h, add 600 μL of Tris-saturated phenol, and fully Shake and mix, 12,000r / min, centrifuge for 10min; take the upper aqueous phase, add an equal volume of phenol: chloroform: isoamyl alcohol (25:24:1) after mixing...

Abstract

The invention discloses a complete nucleotide sequence of a porcine TT virus I CJNY strain and belongs to the field of biotechnology. The length of the sequence is 2,879 bases. The nucleotide homology of three strains of reported TT virus I (GQ120664, AY823990 AND AB076001) with complete genome sequences and the porcine TT virus I CJNY strain is between 68.1 and 86.7 percent. The most different point is that a virus nucleocapsid protein encoded by the sequence only has 415 amino acids which are greatly reduced when compared with those of the other three strains. The sequences can be used for research and development of specific detection technology and is used for research of virus biological characteristics by construction of infectious molecular clone.

Description

technical field [0001] The invention relates to the nucleotide sequence of porcine TT virus type 1 (TTV1) CJNY strain (Anellovirus) and belongs to the field of biotechnology. Background technique [0002] In 1997, Japanese scholar Nishizawa et al. cloned a 500bp fragment N22 from the serum of a patient with non-A-non-G post-transfusion hepatitis using the representative difference analysis technique (Representational difference aIlalysis). Through molecular epidemiological studies, it has been confirmed that N22 is highly correlated with post-transfusion hepatitis. Because the clone was derived from a post-transfusion hepatitis patient named TT, the virus was considered to be related to hepatitis, so it was named TT virus, because TTV and the three prefixes of the English word for transfusion transmitted virus (TTV) The acronym is coincident, so the virus is also known as transfusion-transmitted virus. [0003] TTV is a non-enveloped single-strand circular negative-strand ...
