Problems solved by technology

At present, a lot of work has been done on disease-resistant breeding, chemical control and agricultural measures at home and abroad, but the effect of disease prevention is not satisfactory
And due to the particularity of alfalfa as forage grass and factors such as economy, environment and food safety, there are strict restrictions on the use of chemical pesticides during its growth period.

Embodiment 1

[0022] Example 1. Isolation and identification of strains

[0023] One, separation

[0024] Test soil sample: collected from the rhizosphere soil of healthy alfalfa plants of 2-3 years old in Tongliao, Inner Mongolia.

[0025] Test strains: provided by the Pasture Disease Control Group of the Grassland Research Institute of the Chinese Academy of Agricultural Sciences;

[0026] Medium: ①Seed medium: each liter of water contains 5g peptone, 3g yeast extract, 2g glucose, 1g beef extract, pH 7.0~7.2; ②Basic fermentation medium: each liter water contains 25g soybean meal, 40g starch, 0.8g (NH 4 ) 2 SO 4 , 3g CaCO 3 , 2g NaCl, PH 7.0~7.2.

[0027] Preparation of soil sample dilution: The soil sample was collected from the rhizosphere soil of healthy alfalfa plants of 2-3 years old in Tongliao, Inner Mongolia. When sampling, remove the ground plant residues, use a self-made soil sampler to collect soil samples from 1 to 15 cm, and put them in a sterilized special aluminum dish. The soil sam...

Embodiment 2

[0126] Example 2. Determination of antibacterial spectrum of strains

[0127] The antibacterial spectrum of the strain NMG2-4-8 was determined by the cup and dish method. Put the sterilized Oxford cup into the center of the PDA plate mixed with the target bacteria (plate preparation method: add 1 mL of sterile water into each target bacteria culture test tube, scrape off the colonies, mycelia or spores with an inoculating loop to make a suspension Mix 10 mL of PDA medium and pour it into a 9cm petri dish to make a flat plate), add 200 μL of sterile fermentation filtrate to the cup, and place it at 25°C for 4-5 days (fungus) and 28°C for 2 days (bacteria). The target bacteria will fill the petri dish. The cross method was used to measure the diameter of the bacteriostatic (bacteriolytic) zone. Repeat 3 times for each target bacteria, and average the results.

[0128] PDA agar medium: 200g potato (peeled), 20g sucrose (or glucose), 1000mL distilled water, natural pH.

Abstract

The invention discloses the application of a strain of streptomyces microflavus in alfalfa disease control. The application is particularly the application of streptomyces microflavus NMG2-4-8 CGMCC No.3442 in the biological control of the alfalfa disease. The invention firstly separates the screened one strain of streptomyces microflavus from alfalfa root soil, and an experiment proves that the streptomyces microflavus has an obvious antagonism effect and a wide antimicrobial spectrum. The streptomyces microflavus has strong inhibitory action on crop pathogenic bacteria such as alfalfa downy mildews and the like, and shows good disease resisting effects on alfalfa downy mildew, powdery mildew, tan disease, stemphypium calico, root rot and the like, which indicates that the strain has wide application prospect in the biological control field of alfalfa diseases.

Description

Technical field [0001] The invention relates to the application of a strain of Streptomyces gracilis in the prevention and treatment of alfalfa diseases. Background technique [0002] Biological control of plant diseases emphasizes the virtuous cycle of the ecosystem and environmental protection, which can effectively avoid a series of problems caused by chemical pesticides. It has attracted more and more attention due to its significant social, ecological and long-term benefits. As people pay more and more attention to the concept of ecological balance and green health, biological control measures have received unprecedented attention in agricultural production. Since entering the 21st century, governments and people of various countries have paid more attention to the development and application of biological pesticides, and the share of biological pesticides in the entire pesticide market is increasing year by year. Actinomycetes are the earliest biocontrol microorganisms use...
