Problems solved by technology

These methods use strong acid to dissolve tussah silk fibroin. Due to the strong hydrolysis of strong acid on silk fibroin, a large number of molecular chains of tussah silk fibroin are cut off, and tussah silk peptides or free amino acids with smaller molecular weights are obtained, and larger molecular weights cannot be obtained. tussah fibroin
There are no public reports at home and abroad on the dissolving methods of wild silk fibroin such as celestial silkworm, castor silkworm, willow silkworm, etc.

Embodiment 1

[0020] 1. Remove the silkworm chrysalis in the cocoon of the celestial silkworm, wash the cocoon shell repeatedly with tap water to remove impurities such as dust in the cocoon shell, and then dry the cocoon shell in a 60°C oven to obtain the celestial silkworm cocoon shell after removal of impurities.

[0021] 2. Configure the degumming solution, which contains 0.25% sodium carbonate and 0.1% Marseille soap in the degumming solution, and the rest is water. Put 1 kg of the removed impurities into 50 liters of degumming solution, and treat it twice at 98-100° C. for 60 minutes each time. Then, the cocoon shell of the silkworm is fully cleaned with water, loosened, and dried at room temperature to obtain the degummed silkworm silk fiber.

[0022] 3. Configure a solution containing 9.0mol lithium thiocyanate and 1.0mol ethanol per liter, and the rest is water. Take 1000ml of the solution and place it in a conical flask. Take by weighing 100 grams of degummed celestial silk fibr...

Embodiment 2

[0027] 1. Put 0.2 kg of tussah raw silk into 10 liters of 0.25% sodium carbonate aqueous solution, and process it three times at 98-100°C. Use tap water for the first two treatments, each for 30 minutes. For the third time, use deionized water for 45 minutes. Then, the cocoon shells are fully cleaned with water, loosened, and dried in an oven at 60° C. to obtain degummed tussah silk fibroin fibers.

[0028] 2. Configure tussah silk fiber dissolving solution, each liter of dissolving solution contains 10.0mol calcium nitrate, 1.0mol methanol and 2.0mol propanol, and the rest is water. Take 500ml of the solution and place it in a conical flask. Take by weighing 25 grams of degummed tussah fibroin fiber, cut it into pieces and add it to the conical flask. The Erlenmeyer flask was put into an ultrasonic processor, the ultrasonic power was 1.5kW, and the ultrasonic treatment time was 60 minutes.

Abstract

The invention relates to a preparation for a tussore silk fibroin solution and a concentration method thereof. Fibroin fiber of tussores like tussah, japanese tusser, costor silkworm, and Actias selene Hubner are decontaminated, degummed and then dipped into a dissolving liquid containing neutral salt and alcohol materials, treated by ultrasonic for 5 to 120 minutes, and then dissolved under the temperature between 30 and 98 DEG C to obtain a silk dissolving fibroin. The fibroin dissolving liquid is dialyzed and desalted to obtain a tussore fibroin aqueous liquid; then the aqueous liquid is dewatered and concentrated by a semipermeable membrane by using a high water-absorbing matter to obtain the high concentration tussore fibroin solution with a molecular weight of 15 to 250 kDa and a concentration of 6 to 35 percent. As the neutral salt is used, the large extent hydrolyzation of acid and alkali to the silk fibroin is avoided; therefore, the molecular weight of the obtained tussore silk protein is higher; the tissore silk fibroin can be used for manufacturing tussore silk fibroin gel, powder, films, porous sponges, regenerated fibers, and the like. The invention provides new material for the fields of biomedicine, medicine, make-up, food, health product, and the like.

Description

technical field [0001] The invention relates to a method for preparing and concentrating a natural protein solution, in particular to a method for obtaining a protein solution from silk fibers of wild silkworm silk. The solution can be used to prepare wild silk fibroin protein gel, powder, film, porous sponge, regenerated fiber, etc., and provide new materials for the fields of biomedicine, medicine, cosmetics, food, and health care products. Background technique [0002] Since the silk fibroin obtained from the rear silk glands of cooked silkworms can only meet the needs of a small amount in the laboratory, it cannot meet the requirements of industrial production. Therefore, it is necessary to extract, dissolve and purify silk fibroin fibers to obtain tussah silk fibroin. Protein aqueous solution, and then prepare gel, powder, film, porous sponge, etc. according to the requirements of the application purpose, which are used as raw materials for biomedical materials, medicin...
