Embodiment 1

[0021] Flake dry wakame is made into particles with a diameter of 0.2 mm or less with a super centrifugal pulverizer, and 900 g of powder is suspended and dispersed in 18 kg of water. Then 0.09 g of alginate lyase (from Nagase Biochemical Industries) was added, and the resulting mixture was treated at 45° C. for 18 hours. The resulting product was centrifuged at 5000 rpm for 5 minutes. The precipitate was washed with water, dried, and crushed into samples for protease treatment.

[0022] 25 ml of water in which 1 g of the obtained sample was suspended and dispersed was treated with enzymatic decomposition for 18 hours under a fixed temperature and pH condition after adding 10 mg of each protease. The pH can be adjusted with sodium hydroxide and hydrochloric acid. After the enzymatic decomposition treatment, the product was boiled at 100° C. for 15 minutes to inactivate the enzyme. Centrifuge at 100,000 rpm for 10 minutes to remove the precipitate and obtain a supernatant. ...

Embodiment 2

[0040] Add 2g of dried wakame flakes to 100ml of water to make it swell, and wash with water. Then, 100 ml of water and 20 mg of protease were added, and the mixture was subjected to enzymatic hydrolysis treatment for 18 hours at a fixed temperature and pH value. The pH can be adjusted with sodium hydroxide and hydrochloric acid.

[0041] provided in Table 1

Embodiment 3

[0043] 200g of boiled or salted wakame was added to 3000ml of water and 3mg of alginate lyase, and the resulting mixture was treated at 45°C for 18 hours. After the reaction, the obtained product was centrifuged at 5000 rpm for 5 minutes. The precipitate was washed with water, dried, and crushed to obtain a sample. The treatment per 1 g of the sample was similar to that of Example 1, and 10 mg of each of the four proteases shown in Table 3 was used to determine the ACE inhibitory activity.

[0044] The respective inhibitory activities (%) are Protease S "Amano": 89%; Proleather FG-F: 84%; Protease N "Amano": 75%; and Pepsin: 72%. The dishes are the same.

[0045] Single oral administration (forced administration via gastric probe) of wakame peptides obtained with Proleather FG-F, protease S "Amano" and pepsin induced spontaneously high Blood pressure was administered to male rats (SHR) at a dose of 100 mg / kg, and blood pressure was measured 3, 6, 9 and 24 hours after admini...

Abstract

The object of the invention is to produce a peptide with potent the angiotensin I-converting enzyme (ACE) inhibitory activity by decomposing the protein contained in wakame with protease, using wakame being a food material ingested daily as a raw material, and to provide a safe food composition with hypotensive activity, using the treatment product thereof as a raw material. The inventors have found that, when performing the decomposing treatment of wakame with various proteases, a group of endo-type enzymes produced by Bacillus produce peptides with particularly strong ACE inhibitory activity, leading to the completion of the invention.

Description

technical field [0001] The present invention relates to an angiotensin I-converting enzyme (hereinafter abbreviated as ACE) inhibitory substance capable of lowering blood pressure, and more particularly, to an anti-inflammatory substance obtained by decomposing Wakame (Wakame pinnata, Phaeaceae) with protease. ACE is a peptide with strong inhibitory activity. Background technique [0002] Hypertension is one of the causes of arteriosclerosis, hyperlipidemia, diabetes and obesity. In Japan, it is estimated that 20 million or more people are diagnosed with hypertension, and the number of people with underlying hypertension also reaches a high number. In recent years, high blood pressure is still considered the silent killer, even though the diagnostic criteria for high blood pressure have become stricter. If left unchecked, high blood pressure is asymptomatic but often fatal, so it is considered to be one of the diseases closely linked with lifestyle. In order to improve th...
