[0027] 1. Cell Culture

[0028] Bladder cancer (T24) cells were purchased from ATCC (Manassas, VA, USA) and cultured in Dulbecco's modified Eagle's medium (DMEM) supplemented with 10% fetal bovine serum (FBS) in a 5% CO2 incubator at 37°C .

[0029] The preparation and characteristics of

[0030] The preparation of fiber serpentine has the following different preparation methods: ①Tetraethoxysilane and magnesium chloride hexahydrate are used as raw materials, and MgCl 2 ·6H 2 O was dissolved in deionized water at the ratio of Mg:Si=1:1 to form a solution, and then Si(OCH 3 ) 4 and stirred for 15 minutes to prepare a 0.1M mixed solution; by adding dropwise a 1mol / L NaOH solution and then stirring for 20 minutes, the pH of the solution was adjusted to 12.0; the solution was poured into the reactor for hydrothermal synthesis; synthetic serpentine The samples were obtained by constant temperature reaction at 230°C for 48 hours, followed by repeated...

Abstract

The invention discloses a novel application of a synthetic fiber-like serpentine nano material (SCNT), and particularly relates to an application of the synthetic fiber-like serpentine nano material as a circPRMT5 siRNA carrying material in the aspect of cancer treatment, and further, specific cancer is bladder cancer. According to the invention, a carrier is the synthetic fiber-like serpentine nano material, and siRNA of circular RNA is carried. Experimental results show that the synthetic fiber-like serpentine nano material carrying circPRMT5 siRNA has a good targeting effect on circPRMT5 high-expression bladder cancer cells and tissues, and besides, the synthetic fiber-like serpentine nano material can inhibit growth and metastasis of tumors.

Description

technical field [0001] The invention belongs to the technical field of biomedicine, and specifically relates to the new application of synthetic fiber-like serpentine nanomaterials, specifically as circPRMT5 Application of siRNA delivery materials in cancer therapy. Background technique [0002] Bladder cancer is one of the most common urological malignancies and its prevalence is increasing worldwide. The mortality and incidence of bladder cancer rank first among all urological tumors in China. Although the development of surgical treatment and new combination chemotherapy regimens has improved the median survival to some extent, about 50% of bladder cancer patients develop recurrence / metastasis within 2 years after diagnosis. The high recurrence rate of non-muscle-invasive bladder cancer and the low survival rate of muscle-invasive bladder cancer mean that there is an urgent need for new treatments for bladder cancer. [0003] In recent years, among newly developed can...
