[0045] 1) Preparation of raw materials

[0046] The frozen pig liver was thawed in an ice-water bath, and then the blood on the surface was wiped dry with a paper towel, the film coat and connective tissue were removed and cut into small pieces, and 10 g of pig liver was weighed.

[0047] 2) Preparation of crude enzyme solution

[0048] Add pig liver and 80ml of mannitol buffer solution into a dry and clean tissue homogenizer, mash in the homogenizer at 2000r for 30 seconds, centrifuge the obtained homogenate at 10000r / min for 10 minutes, and take the supernatant 1ml, and finally diluted 10 times with 1% mannitol phosphate buffer solution to prepare crude enzyme solution.

[0049] 3) Determination of catalase activity

[0050] Add 10ml of hydrogen peroxide solution to each of the four iodine measuring bottles and mark them, and place them in a constant temperature water bath at 25°C for 5 minutes. First add 1ml of concentrated sulfuric acid to one of the iodine measuring bo...

Abstract

The invention belongs to the field of enzyme activity detection, and relates to a method for identifying suckling pig liver and adult pig liver by utilizing catalase activity. The catalase is extracted from the pork liver by using an extraction method, the activity of the catalase in the pork liver is determined by using an iodometric method and a potassium permanganate method, and the detection method for identifying the adult pork liver and the suckling pork liver, which is high in accuracy, simple and convenient to operate and high in repeatability, is provided. Experiments show that under the same condition, the catalase activity of the suckling pig liver is obviously smaller than that of the adult pig liver, so that the catalase content in the pig liver can be used as an index for identifying the adult pig liver and the suckling pig liver; the method can fill the blank of methods and standards for detecting adult pig liver and suckling pig liver in China, provides a method which is high in accuracy, simple and convenient to operate and high in repeatability for detection of medicinal suckling pig liver raw materials, and has very high application value.

Description

technical field [0001] The invention relates to a method for distinguishing suckling pig liver and adult pig liver by catalase activity, belonging to the field of enzyme activity detection. Background technique [0002] Catalase widely exists in various tissues of animals and plants, and in most aerobic microorganisms, and is a basic enzyme for scavenging free radicals in animals and plants. The function of catalase is to catalyze the decomposition of intracellular hydrogen peroxide and prevent the oxidative damage of biological macromolecules such as lipids, proteins, and nucleic acids. The current methods of extracting catalase include fermentation and extraction from animal tissues. my country's production of catalase is mainly extracted from animal tissues. This method has the advantages of low operating threshold and abundant sources of raw materials. In terms of economic benefits and difficulty in obtaining raw materials, fresh pork liver is one of the important raw...
