Embodiment 1

[0023] 1. Genomic DNA extraction

[0024] Use the DNA kit to extract DNA samples from bovine liver tissue, and refer to the manual of the kit for specific operation steps; use Quawell-Q5000 ultra-micro spectrophotometer to detect the concentration and purity of the extracted DNA; use 1% agarose gel electrophoresis to detect D260nm / D280nm Integrity of samples with ratios ranging from 1.9-2.1.

[0025] 2. Primer design and synthesis

[0026] According to the bovine OLR1 gene DNA sequence (accession number: NC-037332.1) published in GenBank, exon 1-6 was used as the target, primers were designed using Primer Premier5.0 software, and synthesized by Suzhou Jinweizhi Biotechnology Co., Ltd. The primer sequence is as follows: Table 1 shows:

[0030] 3. PCR amplification and SNP site detection

[0031] Using bovine liver tissue DNA as a template, the first, second, third, fourth, fifth, and sixth exons of the OLR1 gene were ...

Embodiment 2

[0039] 1. Genetic diversity of exon 4 of OLR1 gene

[0040] The frequencies of AA, AG, and GG genotypes of OLR1 gene in grassland red cattle populations were 64.51%, 27.59%, and 6.90%, respectively; the frequencies of A, G alleles were 79.31% and 20.69%, respectively. The AA genotype is the dominant genotype, and the A gene is the dominant allele. The chi-square fitness test showed that the A / G site of grassland red cattle population was in Hardy-Wednberg equilibrium (P>0.05). The genetic homozygosity (Ho) of the OLR1 gene exon 4 polymorphic site in the grassland red cattle population was 0.672, the genetic heterozygosity (He) was 0.328, the effective number of alleles (Ne) was 1.489, and the polymorphic information content (PIC) was 0.274. The calculation results show that the genetic heterozygosity (He) of the fourth exon mutation site of the OLR1 gene is relatively low, indicating that its variation in the grassland red cattle population is small; the polymorphic informat...

Abstract

The invention belongs to the technical field of biology, and particularly relates to an SNP marker related to beef traits of beef cattle and application thereof. According to the SNP marker related to the beef traits of the beef cattle and the application thereof, SNP loci of all exons of ORL1 genes in a grassland red cattle group are detected by using a PCR sequencing method, and the result discovers that only exons 4 of OLR1 genes in the grassland red cattle group have A / G mutation (at Chr5: 99812807bp); chi-square test proves that the SNP loci are in a Hardy-Weinberg equilibrium state in the grassland red cattle group; and genetic analysis discovers that the genetic homozygosity of the genes is 0.672, the genetic heterozygosity is 0.328, the effective allelic factor is 1.489, the polymorphic information content is 0.274, and the genes belong to moderate polymorphism. The correlation analysis result shows that the genotype of the exons 4 of the OLR1 genes has significant difference from the beef traits such as the eye muscle area of the grassland red cattle.

Description

technical field [0001] The invention belongs to the field of biotechnology, and in particular relates to a SNP marker related to meat and beef traits and its application. Background technique [0002] The oxidized low-density lipoprotein receptor OLR1 was first discovered in bovine aortic endothelial cells by Sawamura T et al., and OLR1 was successfully cloned in bovine aortic endothelial cells. The OLR1 gene is mainly involved in the regulation of lipid metabolism in the liver and mammary gland, and is usually expressed at a high level in the lung, liver and adipose tissue, but at a low level in organs such as the heart and ovary. Studies have shown that knocking out the OLR1 gene will inhibit the key enzyme activities that regulate lipogenesis, such as: Acly, Acaca, Fasn and ELOVL6. The bovine OLR1 gene is located on chromosome 5, consisting of 270 amino acids, with a total length of 11306bp, including 6 exons, and has about 72% similarity with human amino acid sequence. ...
