Abstract

The invention discloses application of a macrophage subset and a regulator thereof in acute graft versus host disease (aGVHD). The application of the substance for detecting the expression quantity of a KLF4 transcription factor or a coded nucleic acid of the KLF4 transcription factor in the macrophage comprises the following steps: preparing a product for detecting the polarization / functional state of the macrophage of an aGVHD patient, diagnosing the aGVHD and / or detecting whether M1 / M2 polarization imbalance occurs in the macrophage or not. It is found that significant decrease of the KLF4 in macrophages is a biomarker of M1 / M2 polarization imbalance function change. The macrophage subset and regulator thereof can be used for detecting the polarization and functional state of the macrophage of the aGVHD patient after hematopoietic stem cell transplantation, and has important clinical significance for disease layered diagnosis and treatment.
