Problems solved by technology

[0004] The existing microbial diversity analysis method has errors in sample abundance. The PCR method is used to construct the conventional diversity library. The PCR primers involved are likely to cause deviations due to differences in Tm values, which will cause deviations in the analysis results.

Abstract

The invention discloses an index analysis method based on microbial diversity in selenium-rich soil. The method comprises the following steps of A, weighing a selenium-rich soil sample, and carrying out microbial DNA extraction; B, performing quality detection on extracted metagenome DNA, completing purity and integrity detection of the genome DNAthrough a NanoDrop microspectrophotometer and horizontal agarose gel electrophoresis respectively, and obtaining the DNA mass concentration through Qubit detection; C, performing high-throughput sequencing by adopting a next-generation sequencing technology, and performing library construction on an extracted metagenome sample; and D, performing data analysis: in original data obtained by sequencing, performing quality control on offline data by using FastQC, filtering the data according to a quality inspection report result, accurately estimating the relative abundance of species through rapid comparison, obtaining microbial community composition information, drawing a visual evolutionary tree based on a classification grade by using GraPhlAn, and constructing a species community structure chart.

Description

technical field [0001] The invention relates to the technical field of microbial research, in particular to an analysis method based on microbial diversity index in selenium-enriched soil. Background technique [0002] The community structure and diversity of microorganisms, as well as the function and metabolic mechanism of microorganisms are the research hotspots of microbial ecology. At present, the research on the diversity of soil microorganisms is mainly to obtain the DNA fragments of microorganisms directly from the soil by means of molecular biology. analysis, so that the analysis of environmental microbial diversity becomes a method that is as comprehensive as possible; in recent years, there have been many reports on the methods of microbial diversity analysis in soil, however, the main focus is on the extraction of the total DNA of the bacterial community in the soil and the analysis, less attention has been paid to the extraction and analysis of the total DNA of ...
