Problems solved by technology

Although it is known that this cyclic peptide derivative is extracted from Paecilomyces tenuipes, which is a type of winter grass, and exhibits proliferative activity on astrocytes, it is not known that it acts on small glia and promotes the polarity switch to the anti-inflammatory M2 phenotype of microglia

Embodiment

[0052] 1. Cell Culture of MG6 Microglia

[0053] In the following experiments using a cell line of MG6 microglial cells (Riken Cell Bank), the culture of MG6 microglial cells was performed using Dulbecco's Modified Eagle Medium (DMEM , Thermo Fisher Scientific (thermo Fisher Scientific)). 100 μΜ beta-mercaptoethanol, 10 μg / ml insulin, 10% fetal bovine serum (Gibco), penicillin-streptomycin (Gibco), 450 mg / ml glucose (Gibco) were contained in DMEM.

[0054] 2. Determination of cell viability of MG6 microglial cells

[0055] MG6 microglial cells were cultured in a 96-well plate at 37°C for 24 hours (5×10 3 cells / well), followed by incubation with various concentrations of cyclic peptide derivatives at 37°C for 48 hours. As the cyclic peptide derivative, the diethylamine salt of the compound represented by the above formula (2) obtained by the method described in the Example of Patent Document 2 was used (the same applies to the following tests). The concentration of the cycl...

Abstract

The invention provides a polar conversion accelerator to anti-inflammatory M2 phenotype microglia. The polar conversion accelerator comprises a cyclic peptide derivative represented by formula (1). Inthe formula, m is 0 to 3, n>=1, R1 to R6 are hydrogen atoms or hydrocarbon groups, R7 and R8 are carboxyl or salts thereof or alkoxycarbonyl, R9 is a hydrocarbon group, hydroxyl, alkoxy or alkylcarbonyloxy, R10 and R11 are hydrogen atoms, hydrocarbon groups or alkylcarbonyloxy, and R12 to R16 are hydrogen atoms or hydrocarbon groups. The Chemical formula 1 is shown in the specification.

Description

technical field [0001] The present invention relates to a polarity conversion accelerator capable of promoting the polarity conversion of microglia cells (microglia) to an anti-inflammatory M2 phenotype. Background technique [0002] Microglia, a type of glial cell, can be considered to be cells derived from the mesoderm differentiated from the yolk sac. Microglia are known to function as immune cells that regulate the environment inside and outside the brain, such as removing damaged cells and maintaining and checking synapses. For example, in the case of protecting the brain from traumatic brain injury, microglia activate and release inflammatory cytokines, thereby changing astrocytes into reactive astrocytes, which play a protective role in the brain. In addition, activated microglia are known to induce encephalitis and promote neurodegenerative diseases. [0003] On the other hand, a method for treating neurodegenerative diseases targeting microglia is disclosed. For ...
