preparation example Construction

[0033] According to another aspect of the present invention, a kind of preparation method of the microbial inoculum M2 of preventing and treating corn stalk rot is provided, comprising the following steps:

[0034] (1) Inoculate Bacillus subtilis HR15, Bacillus atrophaeus HR37 and Bacillus veleus HR55 on the slant NA medium respectively, and activate the culture at 30-37°C to obtain Bacillus subtilis HR15 colonies, Bacillus atrophaeus HR37 colonies and Belais Bacillus sp. HR55 colonies, the slant NA medium is a solid medium.

[0035] (2) Inoculate the activated Bacillus subtilis HR15 colony, Bacillus atrophaeus HR37 colony and Bacillus Veles HR55 colony into NA liquid medium respectively, under the condition of 30~37℃, 180~220rpm / min Shake culture for 10-15 hours to obtain the seed fermentation liquid of Bacillus subtilis HR15, the seed fermentation liquid of Bacillus atrophaeus HR37 and the seed fermentation liquid of Bacillus Velez HR55.

[0036] (3) Bacillus subtilis HR15 ...

Embodiment 1

[0045] The determination of compatibility between the strains of embodiment 1

[0046] Referring to the determination method of Barbosa, one functional strain was streaked on half of the LB solid medium plate, and another functional strain was vertically streaked on the other half, and each treatment was repeated 3 times. Place the plate in a biochemical incubator at 37°C for cultivation, and observe and record the compatibility reaction between the two strains after 24 hours. The two strains that do not produce antibacterial bands are compatible, otherwise they are incompatible. Compatibility results see figure 1 .

[0047] LB solid medium: 5g of yeast extract, 10g of peptone, 5g of sodium chloride, 1000mL of water, pH7.4-7.6, 15-20g of agar powder, sterilized at 121°C for 30min.

Embodiment 2

[0048] The preparation of embodiment 2 microbial bacterial agents M2 powder

[0049] Incline strains: using solid NA medium, inoculate Bacillus subtilis HR15, Bacillus atrophaeus HR37 and Bacillus velez HR55 on the slant NA medium respectively, and culture them in an incubator at 37°C for 1-2 days.

[0050] Meat juice peptone liquid medium (NA): 0.3% beef extract, 0.7% peptone, 0.3% NaCl, pH7.2. Bouy peptone solid medium (NA): add 15-20g agar powder on the basis of liquid medium to obtain solid medium.

[0051] Preparation of Bacillus fermentation broth: inoculate single colonies of strains HR15, HR37 and HR55 activated for 24 hours into seed fermenters equipped with liquid NA medium, and vibrate at 200 rpm / min for 12 hours at 37°C to obtain seed fermentation liquid. The seed fermentation broths of the three strains were respectively transferred to fermentors at a ratio of 6%, and cultured with shaking at 200 rpm / min for 48 hours at 37°C.

Abstract

The invention discloses a microbial agent M2 for preventing and treating corn stalk foot rot and a preparation method therefor. The microbial agent M2 is prepared from a mixed bacterial solution, a support, humic acid, zinc sulfate, ferrous sulfate and potassium sulfate, wherein the mixed bacterial solution is prepared through blending bacillus subtilis HR15, bacillus atrophaeus HR37 and bacillusvelezensis HR55 according to a volume ratio of 1: 1: 1, and a chicken-manure organic fertilizer serves as the support. The three kinds of strains are all collected in China General Microbiological Culture Collection Center of China Committee for Culture Collection of Microorganisms on March, fifth, 2020. The bacillus subtilis HR15 has an accession number of CGMCC NO. 19453; the bacillus atrophaeusHR37 has an accession number of CGMCC NO. 19451; and the bacillus velezensis HR55 has an accession number of CGMCC NO. 19452. The microbial agent M2 disclosed by the invention has a good prevention effect on the corn stalk foot rot and is remarkable in yield increasing and growth promoting effects.

Description

technical field [0001] The invention relates to the technical field of microorganisms, in particular to a microbial agent M2 used for the prevention and treatment of corn stalk rot and a preparation method thereof. Background technique [0002] Corn is an important food crop in the world, and its planting scale ranks only after wheat. With the change of global climate conditions, continuous cropping, high-density cultivation, etc., the ecological environment of cornfields has changed accordingly, the structure of the dominant flora of pathogenic bacteria in the soil has changed, the accumulation of diseased residues has increased, and the structure of beneficial microbial communities and quantities have also changed significantly. These factors lead to the imbalance of soil microbial community structure in cornfields, and the occurrence of corn stalk rot is becoming more and more serious. [0003] Maize stalk rot is a typical soil-borne disease caused by complex infection ...
