Problems solved by technology

However, complex temperature control procedures and a single sample injection method are still the core issues restricting the development and renewal of qPCR devices
Most commercially available real-time qPCR platforms still require frequent temperature control operations, which increases the time spent on nucleic acid amplification and detection and reduces reaction efficiency
However, in the operation process of traditional qPCR equipment, multiple samples must be reacted in batches, tested uniformly, and ended at the same time, which makes it difficult for traditional qPCR equipment to achieve flexible arrangement of single or different batches of samples, so that it cannot become a real high-throughput detection

Abstract

The invention relates to a device, a system and a method for real-time fluorescence quantitative nucleic acid amplification detection. The device comprises a micro-fluidic unit, a sample injection unit, a temperature control unit and a driving unit; the micro-fluidic unit is provided with one or more parallel annular flow channels; the annular flow channels are provided with a liquid inlet and a liquid outlet; the sample injection unit is arranged at the liquid inlet of the annular flow channels and is used for injecting samples into the annular flow channels; the temperature control unit is matched with the micro-fluidic unit; the driving unit is matched with the micro-fluidic unit and is used for driving a micro-fluid in the annular flow channels to move; at least one section of the annular flow channels is made of a transparent material, so that fluorescence detection is facilitated; and different flow sections of the annular flow channel have different preset temperatures through configuration of the temperature control unit, and through moving in the flow sections with different temperatures in the annular flow channels, the micro-fluid realizes temperature change operation. The invention simplifies nucleic acid amplification and detection operation, is flexible to control, reduces detection time and material cost, and improves detection efficiency.

Description

technical field [0001] The invention relates to the technical field of nucleic acid amplification detection, in particular to a device, system and method for real-time fluorescent quantitative nucleic acid amplification detection. Background technique [0002] Polymerase chain reaction (PCR) is a technique widely used in molecular biology. By simulating the DNA replication conditions in organisms, a DNA polymerase reaction is used to specifically amplify a certain DNA fragment. PCR technology is essential for gene sequencing, molecular cloning and gene mutation analysis in human diagnostics. The PCR reaction process usually involves three steps of denaturation, annealing, and extension, and these steps need to be realized by changing the temperature of the reaction system and controlling the temperature. A PCR machine or thermal cycler is used for the amplification and detection of nucleic acid samples by adjusting the temperature during the cycle program. According to the...
