Abstract

The invention discloses a method for purifying kerriin A from Pittosporum kerrii craib velamen by column chromatography-high speed countercurrent chromatography. The method comprises the following steps: step A, taking a proper amount of Pittosporum kerrii craib velamen powder, carrying out heat reflux extraction with an ethanol aqueous solution, filtering and concentrating an extracted solution until no alcohol smell exists, carrying out isopyknic extraction with ethyl acetate, and concentrating an obtained ethyl acetate fraction under reduced pressure to obtain an ethyl acetate extract; stepB, dissolving the ethyl acetate extract in a proper amount of an ethanol aqueous solution; filtering with absorbent cotton, loading a sample to a D101 macroporous adsorption resin column, eluting with 10 column volume times of an ethanol aqueous solution with the volume percentage concentration of 30% to remove impurities, eluting with an ethanol aqueous solution with the volume percentage concentration of 65%, collecting a sixth column volume eluent, recovering ethanol under reduced pressure, and carrying out freeze drying to obtain a kerriin A enriched product; and step C, purifying the kerriin A by high speed countercurrent chromatography.
