Abstract

The invention relates to the technical field of tissue culture, in particular to a directional culture medium for germinated orchid seeds and a method for directional cultivation of seedlings. The directional medium includes, proliferation medium: MS medium containing sucrose, agar powder, 6-BA, NAA; differentiation medium: MS medium containing sucrose, agar powder, KT, IBA; rooting medium: containing sucrose , agar powder, 6-BA, IAA MS medium. In this application, the germinated seeds of Shanlan were used as test materials to study the regulation and promotion of different mitogens and auxins and their ratios on the process of Shanlan from germination to seedling development. The results show that 6-BA and NAA are beneficial to protocorms. Proliferation, KT and IBA are conducive to the differentiation of protocorms into corms, and 6‑BA and IAA are conducive to corm rooting. The method of the invention enables the orchid germinated seeds to develop directionally, shortens the time for seedling formation, and improves the efficiency of seedling formation.
