The device comprises a negative-pressure sample feeding pump and a microflow control chip, wherein the microflow control chip comprises an upper-layer sample feeding layer, a plastic micro-hole filtering film and a lower-layer waste liquid layer; a plurality of filtering holes are uniformly formed in theplastic micro-hole filtering film; the upper-layer sample feeding layer is communicated to a sample test tube; and the lower-layer waste liquid layer is connected to a negative-pressure feeding pump.The invention further provides a preparation process of the filtration and microflow control device for the circulating tumor cells. The preparation process comprises the following steps: preparing the upper-layer sample feeding layer and the lower-layer waste liquid layer, preparing the plastic micro-hole filtering film, assembling the microflow control chip and carrying out integral mounting. The invention further provides a working method of the filtration and microflow control device. The working method comprises the steps of preparing a sample, capturing tumor cells, adding an antibody diluent and carrying out immunofluorescent staining observation and counting.
